悬吊大鼠胃窦部和十二指肠5-HT免疫反应细胞的变化

Alterations of 5-HT Immunoreactive Cells in The Gastric Antrum and Duodenum in Tail Suspended Rats

摘要 为了探讨失重因素对胃肠5-羟色胺免疫反应(5-HT-IR)细胞的影响,我们用免疫组织化学ABC 法,对地面模拟失重大鼠和正常对照大鼠胃窦部和十二指肠的5-HT-IR 细胞进行了观察。结果表明,模拟失重大鼠胃幽门部及十二指肠各段的5-HT-IR 细胞均较对照大鼠明显增多。本文对这一结果及其意义进行了初步讨论。 Weightlessness has been noted to have widespread effects on bodyfunctions.Yet no study on the response of endocrine functions of gastrointestinal tract toweightlessness has been reported.Serotonin,an important bioaetive substance,which ex-tensively regulates the functions of the body is mainly produced in the EC cells of the gas-trointestinal tract.The purpose of this study was to reveal the alterations of 5-HT im-munoreactive cells(5-HT-IR cells,i.e.EC cells)in rats after 90 d of simulated weight-lessness produced by tail suspension.Altogether 16 male Wistar rats(30 days old)wereused.They were divided into two groups randomly and equally,one group for ground-based weightlessness simulation and another for normal control.The rats used for ground-based weightlessness simulation were suspended by the tail in head-down position(-30°)with hind limbs bearings no weight for three months.Food and water were suppliedfreely.The control rats were fed with the same water and food in the same room exceptsuspension.Animals were sacrificed by over dose pentobarbital.The gastric antra andduodena were taken out and fixed in paraformaldehyde for 24h and then embeded rou- tinely in paraffin.Sections of 5 μm were made.ABC immunostaining procedure wasadopted to reveal the 5-HT-IR cells.The results showed that the shapes of the gut 5-HT-IR cells in simulated weightlessness rats were flask-like,triangular or basket-like with orwithout apical processes,which showed no difference to the EC cells in the control rats.However,the densities of 5-HT-IR cells in the gastric antrum,middle and lower part ofduodenum of the simulated weightlessness rats were increased by 42.0%,46.2% and40.7% respectively as withmpared to the counter parts of the control rats(P<0. 01).In the upper part of duodenum,5-HT-IR cells were increased by 25.0%(P<0. 05).The degrees of enhancement in different parts were not significantly different.Althoughthe increases of the 5-HT-IR cells do not necessarily mean increases of synthesis or releaseof 5-HT,For the decrease of release may also give rise to the same result,however,onthe condition of long periods of simulated weightlessness,we inclined to the enhancementof 5-HT.for the drop of release would result in cut down of enzymes for the synthesis of5-HT.Anyway,the present result implies that 5-HT might play a role during weightless-ness.What the patho-physiological meaning is of the increases of 5-HT-IR cells is fasci-nating and awaits further investigation.
