摘要
目的观察重组人胰高血糖素样肽 1[rhGLP 1(7 36 )NH2 和rhGLP 1(7 37) ]对实验性糖尿病大鼠模型血糖和血浆胰岛素水平的影响。方法正常SD大鼠按 3.2 4g/kg腹腔注射 5 0 %葡萄糖 ,诱发暂时性高血糖动物模型。腹腔注射不同剂量 (2 0、4 0、80 μg/kg)rhGLP 1(7 36 )NH2 和rhGLP 1(7 37) ,间隔取血测定血糖与血浆胰岛素水平。正常SD大鼠按 6 0mg/kg腹腔注射链脲佐菌素 ,建立实验性 1型糖尿病大鼠模型。静脉输注rhGLP 1(7 36 )NH2 [4.0ng/(kg·min) ]12 0min并观察血糖变化。结果腹腔注射 4 0、80 μg/kgrhGLP 1(7 36 )NH2 和rhGLP 1(7 37)处理组大鼠的血糖、血浆胰岛素水平与对照组之间的差别具有非常显著性意义 (P <0 .0 0 1) ,且呈现一定的量效关系。二者之间促胰岛素分泌和降血糖活性差异无显著性。静脉输注rhGLP 1(7 36 )NH2 6 0min后 ,血糖水平 (17.0 4± 1.31)mmol/L(P <0 .0 5 ) ,12 0min后血糖水平 (11.98± 1.0 5 )mmol/L(P <0 .0 0 1)。结论rhGLP 1能显著改善实验性糖尿病大鼠的血糖水平 ,这与其促胰岛素分泌活性有关。
PurposeTo study the effects of two recombinant human glucagon-like peptide-1 [rhGLP-1(7- 36)NH 2 and rhGLP-1(7-37)] on plasma glucose and insulin in the experimental diabetic SD rats.MethodsSD rat model with hyperglycemia was developed by intraperitoneal injection in these normal SD rats with 50%glucose 3.24 g/kg. rhGLP-1(7-36)NH 2 and rhGLP-1(7-37) were administered at different doses, then blood samples were collected and the levels of plasma glucose and insulin were determined. Subsequently, the experimental 1 model diabetic rats were produced with streptozotocin on 60 mg/kg. rhGLP-1(7-36)NH 2 [4.0 ng/(kg·min)] was intravenously injected into the diabetic rats for 120 min and the levels plasma glucose was measured.ResultsCompared with glucose group, plasma glucose levels were significantly decreased after administration 40,80 μg/kg rhGLP-1(7-36)NH 2 and rhGLP-1(7-37) in the experimental SD rat model. Besides, the levels of insulin in plasma were also apparently increased in these rats as compared with glucose rats (P<0.001). Both rhGLP-1(7-36)NH 2 and rhGLP-1(7-37) could improve the levels of plasma glucose and insulin, but these indexes showed no significant difference between them. In the type 1 diabetic rats, plasma glucose concentrations were(17.04±1.31) mmol/L (P<0.05) after 60 min by intravenous injection rhGLP-1(7-36)NH 2, and glucose concentrations were(11.98±1.05) mmol/L (P<0.001) after 120 min.ConclusionrhGLP-1 treatment could improve plasma glucose levels in the experimental diabetic rats model. It might be related to stimulating the secretion of plasma insulin in the rats model.
