哈尼草莓叶片诱导再生植株及快速繁殖研究 被引量:2

Induction of Regenerated Plantlet From Leaf Disc and Micropropagationof Honeoye Strawberry (Fragaria)

摘要 以哈尼草莓组培苗叶片为材料,建立了一个诱导再生植株的高效培养体系。以茎尖为外植体,以MS为基本培养基研究了不同浓度和配比的6-苄基腺嘌呤(6-BA)、奈乙酸(NAA)和二氯苯氧乙酸(2,4-D)对芽增殖和生根的影响,筛选出芽增殖、生根的较适宜培养基。结果表明:①叶片愈伤组织的诱导以MS+6-BA0.1mg/L+2,4D0.1~0.5mg/L较好,愈伤率达100%;②愈伤组织在MS+BA0.5mg/L+IBA0.01mg/L培养基上,芽再生频率较高,平均再生芽数为3.2;③芽增殖以MS+6-BA0.5mg/L+IBA0.05mg/L效果好,增殖系数为9.8;④生根较适宜培养基为1/2MS+NAA0.3mg/L,生根率达100%,平均每株生根数11条,且根生长粗壮。 An efficient protocol had been developed for shoot organogenesis from the leaf disc of vitro-cloned Honeoye strawberry plantlet and the effects of different concentrations and combinations of benzylaminopurine (BA), alpha-naphthaleneacetic acid (NAA) and 2,4-dichlorophenoxy acetic acid (2,4-D) on shoot multiplication and rooting were investigated. The results showed that ①the better media for the induction of callus from leaf disc were +BA 0.1mg/L+2,4-D 0.1-0.5mg/L, callus rate were 100%; ②the highest shoot regeneration frequency and an average of 3.2, shoots per explant were obtained on MS+BA 0.5mg/L+IBA 0.1mg/L; ③MS+BA 0.5mg/L+IBA 0.05mg/L was the suitable medium for shoot multiplication(9.8 shoots per explant); ④shoots were rooted on 1/2MS+NAA 0.3mg/L with rooting rate 100% and per shoot regenerated eleven roots.
