腺病毒介导MnSOD转染对肺癌细胞生长及体内抗肿瘤作用的影响 被引量:9

摘要 目的研究腺病毒介导锰超氧化物歧化酶(Ad-MnSOD)对肺腺癌细胞生长影响及在体内抗肿瘤作用。方法应用细胞基因转染、四甲基偶氮唑蓝(MTT)比色、逆转录聚合酶链反应(RT-PCR),观察Ad-Mn-SOD转染A549细胞后对肺癌A549细胞生长的影响,并将A549细胞接种到裸鼠皮下后,注射Ad-MnSOD及Ad-LacZ到瘤体内,观察对肿瘤生长的影响。结果50MOI Ad-Laze转染率达90%以上,Ad-MnSOD转染A549细胞后MnSOD mRNA表达增加,MTT法检测显示转染Ad-MnSOD A549细胞组生长率(78.7%)低于转染Ad-LacZ的对照组(92.2%);A549细胞接种裸鼠后,用Ad-MnSOD及Ad-LacZ分别注射于瘤体内,Ad-MnSOD组瘤体生长速度低于Ad-LacZ对照组。结论Ad-MnSOD转染A549细胞后MnSOD可明显抑制A549细胞生长及肿瘤在裸鼠体内的生长,MnSOD对肺癌细胞A549有明显抗肿瘤作用。 Objective: To study the biological action of tumor cells infected with adenovirus-mediated MnSOD and determinate the antitumor effect of Ad-MnSOD. Methods: Lung adenocarcinoma cell line A549 was transfected by Adenovirus-mediated MnSOD gene. Cell growth inhibition assay was used to observe the effect on tumor cells. The expression of MnSOD mRNA, which was transduced into A549 cells mediated by denovirvs, was detected by RT-PCR. The intratumoral injection of Ad-LacZ and Ad-MnSOD was carried out to evaluate their antitumor effects. Results: More than 90% of A549 cells could be infected by 50 multiplicity of infection (MOI) Adenovirus. The expression of MnSOD mRNA in A549 cell transfected by Ad-MnSOD increased greatly. The A549 cells transduced with Ad-MnSOD grew slower than those transduced with Ad-LacZ. Antitumor study indicated that tumor bearing mice treated with Ad-MnSOD also grew slower than the controls which treated with Ad-LacZ, and the tumor volume was significantly smaller than that of the controls. Conclusion: The growth of tumoral cells A549 are suppressed after infected with Adenovirus-mediated MnSOD. The intratumoral injection of Ad-MnSOD cas inhibite the growth of solid tumor. MnSOD demonstrates the effect of antitumor.
