摘要
目的 :观察七叶亭对实验性骨关节炎关节液中一氧化氮 (NO)、前列腺素E2 (PGE2 )和软骨中基质金属蛋白酶 1(MMP 1)水平的影响 ,探讨七叶亭对骨关节炎的修复作用。方法 :2 4只日本大耳白兔随机分成对照组A、B和治疗组C、D。对照组A只行单侧膝关节切开术作假手术对照 ,对照组B及治疗组C、D均行单侧前交叉韧带切断术建立骨关节炎模型。治疗组C术后行小剂量 10 0mg·kg-1七叶亭腹腔注射 ,治疗组D术后行大剂量 2 0 0mg·kg-1七叶亭腹腔注射 ,对照组均给予生理盐水 ;8周处死 ,抽取关节液作NO、PGE2 测定 ,以免疫组织化学方法检测软骨中MMP 1的表达 ;对标本均行大体观察及光镜观察。结果 :治疗组软骨退变明显轻于对照组B ,MMP 1、NO、PGE2 含量明显低于对照组B ,治疗组D尤为明显。结论 :七叶亭能明显降低关节炎关节软骨中的MMP 1及关节液中的NO、PGE2 水平 ,减缓骨关节炎的发生。
Objective: To research the effect of esculetin on nitric oxide, prostaglandin E 2 of synovial fluid and matrix metalloproteinase-1 (MMP-1)in cartilage in the model of experimental knee osteoarthritis and to determine the effect of esculetin in repairment of osteoarthritis. Methods: 24 rabbits were divided into control group A, B and treatment group C, D at random. Osteoarthritis was induced in 18 rabbits of group B, C and D by anterior cruciate ligament-transection. The other 6 rabbits (control group A) received unilateral knee arthrotomy as pseudo operative controls. Treatment group C were injected intraperitoneally with esculetin 100 mg·kg -1 ·d -1 after operation. Treatment group D were injected intraperitoneally with esculein 200 mg·kg -1 ·d -1 after operation. Control group B received normal saline. All rabbits were killed at 8 weeks post-surgery, synovial fluid were collected, and NO and PGE 2 were examined. The distribution of MMP-1 of cartilage was documented by immunohistochemistry. Samples of joint were collected for general and light microscopic observation. Results: Cartilage degeneration in treatment group was significantly slighter than that in control group B. The levels of MMP-1,NO and PGE 2 in treatment group were lower significantly than those in control group B, expecially in treatment group D. Conclusion: Esculetin can significantly reduce the expression of MMP-1 in cartilage and levels of NO and PGE 2 in synovial fluid, and postpone the occurring of osteoarthritis.
