摘要
目的 :观察大鼠肝癌模型腹腔阿霉素化疗引起的心脏氧化损伤以及复合抗氧化剂的保护作用 .方法 :采用Walk er 2 5 6肿瘤细胞株接种大鼠皮下 ,得到实体瘤 ,将实体瘤分割成小块植入大鼠肝脏 ,制成大鼠移植性肝癌模型 ,将肝癌模型大鼠随机分成 3组 ,分别为模型组、单纯化疗组和抗氧化剂组 ,同时选取经手术但未接种肿瘤的大鼠作为阴性对照组 .抗氧化剂组每日给予复合抗氧化剂灌胃 ,单纯化疗组和抗氧化剂组每 2d进行一次腹腔阿霉素化疗 ,剂量为 2mg/kg ,8wk后处死大鼠 ,取心脏制成匀浆 ,分别测定丙二醛 (MDA)、总超氧化物歧化酶 (T SOD)、总抗氧化力 (TAC)和总蛋白含量 .结果 :单纯化疗组的MDA含量较阴性对照组和模型组显著升高 (P <0 .0 1 ) ,而抗氧化剂组的MDA含量较单纯化疗组显著降低 (P <0 .0 1 ) .单纯化疗组T SOD活性和TAC非化疗组显著降低 (P <0 .0 1 ,P <0 .0 5 ) ,抗氧化剂组T SOD活性和TAC较单纯化疗组显著升高 (P <0 .0 1 ,P <0 .0 5 ) .结论 :大鼠肝癌腹腔阿霉素化疗可以引起心脏的氧化损伤 。
AIM: To investigate the oxidative heart injury of rats with liver embedded tumor induced by adriamycin and the protective effects of multiple antioxidants on it. METHODS: The tumor cells (Walker 256) were injected into the rats to create the entity tumors. When the entity tumors were removed from the rats, they were cut into small pieces and embedded into the rats’ livers. The rats with entity tumor were randomly divided into three groups: Tumor group, chemotherapy group and protective group. The rats without liver embedded tumor were selected as negative controls. The protective group was given compound antioxidants by gavage daily and the tumor group and chemotherapy group were given adriamycin by intraperitoneal injection at the dosage of 2 mg/kg body weight every two days. All the rats were killed at the end of eighth week. After their hearts were collected,the content of malondialdehyde (MDA), the activities of total superoxide dismutase (T SOD) , the total antioxidant capacity(TAC) and the total proteins were measured. RESULTS: The content of MDA in the chemotherapy group was much higher than that in the control group and tumor group( P <0.01) but the content of MDA in the protective group was obviously lower than that in the chemotherapy group( P <0.01). Compared with that in the other groups, the activity of T SOD and TAC in the chemotherapy group decreased greatly ( P <0.01, P < 0.05) . Compared with that of the chemotherapy group, the activity of T SOD and TAC in the protective group increased significantly( P <0.01, P <0.05). CONCLUSION: The chemotherapy of liver embedded tumors with adriamycin can cause oxidative injury to the hearts of rats but multiple antioxidants have some protective effect on the injury.
