摘要
目的 :建立生物材料中异烟肼的高效液相色谱快速分析方法 ,研究异烟肼在中毒家兔体内的分布。方法 :以香草醛为衍生化试剂 ,经柱前衍生为异烟肼 香草醛腙 ,直接对体液和组织中的样品进行定性、定量分析。结果 :体液和组织在所建方法中的线性范围是 0 .2~ 1 2 .0mg·L-1 和 0 .5~ 1 2 .0 μg·g-1 ;检测限为 0 .2mg·L-1 和 0 .5 μg·g-1 ;日内、日间精密度值小于4 .9%和 5 .1 %;回收率在 97.1 %~ 1 0 1 .3%和 95 .5 %~ 1 0 3.3%之间。结论 :异烟肼在一次性急性中毒死亡家兔体内分布依次减小的顺序为尿、胃、肾、脑脊液、玻璃体、肺、脑、血、心、肝。该分析方法适应于法医学鉴定、毒代动力学和分析毒理学等相关研究。
OBJECTIVE For the purpose of getting data and analysis of the distribution of isoniazid in rabbit with intoxication a reversed-phase HPLC was developed. METHODS Vanillin, being as a derivative reagent, was added to rabbit's body fluid and tissues when they are pretreated. Isoniazid and vanillin reacted to form isonicotinoyl hydrazone which was separated and detected. RESULTS The linear range was 0.2~ 12.0 mg·L -1 and 0.5~ 12.0 μg·g -1, the limit of detection was 0.2 mg·L -1 and 0.5 μg·g -1. The intra and inter-day precision of assay for isoniazid was less than 4.9% and 5.1% respectively. The average recoveries of isoniazid were from 97.1% to 101.3% in body fluid and from 95.5% to 103.3% in tissues. The HPLC method has been used to investigate the concentration of isoniazid in intoxication rabbit's body fluid and visceras. CONCLUSION The concentration of isoniazid was decreasing in acute poisoning rabbit's urine, stomach, kidney, spinal fluid, vitreum, lung, brain, blood, cardia, liver in order. The quantitative analysis of isoniazid in samples was not interfered by inner impurity matters, the metabolic matters of isoniazid and the derivative reagent residue. The HPLC method for isoniazid is sensitive, rapid and convenient. It is suitable for the analysis of toxic agent in forensic medicine, the detection of drug concentration in the clinical medicine and other studies.
