摘要
目的:运用X?Y双色探针行人类胚胎分裂球植入前诊断, 探讨荧光原位杂交技术在人类胚胎植入前诊断中的应用价值?方法:对常规获得的胚胎, 采用胚胎活检或直接行胚胎透明带的溶解,获取胚胎分裂球采用X?Y双色着丝粒探针进行荧光原位杂交(fluorescent in situ hybridization, FISH)分析?结果:FISH分析成功率为79.49%,有价值结果获得率为23.98%?采用活检方法固定率?杂交率?完整信号率和有价值结果获得率分别为84.06%?77.59%?33.33%?21.74%;直接消化透明带固定率?杂交率?完整信号率和有价值结果获得率分别为77.17%?80.61%?40.51%?25.20%?两者相比无明显差异?结论:X?Y双色探针荧光原位杂交技术适用于胚胎植入前单个胚胎分裂球的遗传学诊断?
Objective: To study preimplantation of embryo blastomeres with X, Y dual color probes, and to search the clinical value of fluorescent in situ hybridization (FISH) in preimplantation diagnosis. Methods: Embryos biopsy were perfomed or all the oocyte zone was dissolved, single cells were harvested and followed by fixation and in situ hybridization. Results: The ratio of hybridization was 79.49%, the ratio of valuable results obtaining was 23.98%. With the method of biopsy, the ratios of fixation, hybridization, fluorescent signals after hybridization and full fluorescent signal were 84.06%, 77.59%, 33.33% and 21.74%, respectively which had no significant differences compare with the methods that all the oocyte zone was dissolved and shaped single cells were got directly (77.17%, 80.61%, 40.51% and 25.20%). Conclusion: Fluorescent in situ hybridization with X,Y dualcolr probes is valuable for genetic diagnosis before implantation. [
