谷胱甘肽转硫酶M1、T1基因型及烟酒嗜好与胃癌易感性的关系 被引量:6

Relationship Between Glutathione S-Transferase M1,T1 Genetic Polymorphisms, Smoking and Alcohol Consumption and Susceptibility to Stomach Cancer

摘要 目的探讨谷胱甘肽转硫酶M1(GSTM1)、T1(GSTT1)基因多态性及烟酒嗜好与胃癌易感性之间的关系。方法采用1∶1配对病例-对照研究方法和分子流行病学研究技术检测60例原发性胃癌患者和60例健康对照的GSTM1和GSTT1基因型,结合研究对象的烟酒嗜好,分析其在胃癌发生中的作用及基因与烟酒嗜好间的交互作用。结果GSTM1(-)基因型出现频率在病例组和对照组中分布差异无显著性,GSTM1-与胃癌易感性无关联(χ2=1.63,P>0.05);GSTT1(-)基因型出现频率在病例组和对照组中分布差异有显著性,GSTT1-基因型增加胃癌发病危险(χ2=6.08,P<0.05,OR=2.8395%可信区间=1.24~6.47);饮酒者较非饮酒者发生胃癌的危险高(OR=2.51,95%可信区间=1.09~5.95)。同时携带GSTM1(-)和GSTT1(-)基因型者发生胃癌的危险性高于GSTM1(+)和GSTT1(+)基因型携带者(OR=3.27,95%可信区间=1.24~8.54,P<0.05),携带GSTM1(-)和GSTT1(-)且有烟酒嗜好者,发生胃癌的危险显著高于GSTM1(+)和GSTT1(+)的无烟酒嗜好者(OR=12.00,95%可信区间=1.99~72.36。结论GSTT1(-)基因型、饮酒可增加胃癌易感性,GSTM1和GSTT1基因型与烟酒嗜好在胃癌发生中存在一定的交互作用。 Objective To determine the association of glutathione-stransferase M1(GSTM1), T1(GSTT1) genetic polymorphisms, smoking and alcohol consumption with the risk of gastric cancer. Methods 1:1 matched case-control study and molecular epidemiology technique were used to analysis the genetic polymorphisms of GSTM1, GSTT1 genotype among 60 cases of gastric cancer patients and sex-and age-matched controls, who were selected from Nanjing Zhongda Hospital and Jiangsu Province Cancer Hospital. Statistical analysis was performed to elucidate interaction of GSTM1. GSTT1 genotype and smoking and alchol consumption in the risk for stomach cancer. Results The frequency of GSTT1 null genotype among cases and controls was 61.67% and 40.00% respectively (χ2 =6.08, P<0.05), which was significantly associated with stomach cancer, OR=2.83 (95%CI =1.24-6.47);Alcohol consumption was significantly associated with stomach cancer, OR=2.51(95%CI=1.06-5.95) GSTM1 null genotype was more commonly observed in the cases, but this was not statistically significant;the same with cigarette smoking. An interaction was observed between GSTM1, GSTT1 genetic polymorphisms and smoking and alcohol consumption to stomach cancer. People who carried GSTM1(-), GSTT1(-) and smoking and alcohol consumption had a higher susceptibility to stomach cancer than those with GSTM1(+), GSTT1(+) and non-smoking and alcohol consumption, (OR=12.00 95%CI =1.99-72.36). Conclusion GSTT1 null genotype and drinking habit might be likely associated with the susceptibility to stomach cancer; interaction of GSTM1?GSTT1 genotype and smoking and alcohol consumption existed in the risk of stomach cancer to a certain extent.
