红霉素对大鼠下丘脑外侧区葡萄糖敏感神经元的作用

EFFECTS OF ERYTHROMYCIN ON THE GLUCOSE-SENSITIVE NEURONS IN THE LATERAL HYPOTHALAMIC AREA IN RATS

摘要 ①目的 探讨大鼠下丘脑外侧区 (LHA)葡萄糖敏感神经元 (GSNs)是否参与了中枢胃动素对摄食行为的调控。②方法 应用细胞外记录神经元单位放电的方法 ,记录麻醉大鼠 (n =32 )LHA的神经元电活动。左颈总动脉注射 0 .5 6mol/L的葡萄糖溶液 0 .2mL ,以 0 .2 8mol/L的高渗盐水溶液作对照 ,放电频率降低者为GSNs,其余为非葡萄糖敏感神经元 (NGSNs) ;侧脑室注射 4 g/L的红霉素 1 μL ,观察其对GSNs及NGSNs的自发放电频率的影响 ,以同等剂量的生理盐水溶液作对照。③结果 在LHA记录到的 4 5个电活动神经元中 ,GSNs 1 8个 ,NGSNs 2 7个 ;侧脑室给予红霉素后 ,在 1 8个GSNs中 ,有 1 2个 (6 6 .7% )放电频率增加 ,3个放电频率减少 ,其余无反应 ;在 2 7个NGSNs中有 7个 (2 5 .9% )表现为兴奋 ,5个表现为抑制 ,其余无反应。红霉素对GSNs和NGSNs的作用差异有统计学意义 (χ2 =8.34,P <0 .0 5 )。④结论 胃动素受体激动剂红霉素对LHA的GSNs有明显的兴奋作用 。 Objective To explore the involvement of glucose-sensitive neurons (GSNs) in lateral hypothalamic area (LHA) in central motilin-induced feeding behavior. Methods Extracellular recordings were made from single neuron in LHA in anesthetized rats. According to their responsiveness to intracarotid injection of glucose solution(0.56 mol/L) 0.2 mL, the neurons in LHA with decreased firing frequencies were classified as GSNs and the others were non-glucose-sensitive neurons (NGSNs). 0.28 mol/L NaCl solution was used to the controls. Effects of i.c.v. administration of 4 g/L erythromycin 1 μL on neuronal activities of GSNs and NGSNs were examined and normal saline was used as control. Results Eighteen GSNs and 27 NGSNs were recognized in LHA. After erythromycin was microinjected by i.c.v., the firing rates increased in 12 GSNs and decreased in three GSNs , but the rest had no changes. Seven neurons were excited and five neurons were inhibited in 27 NGSNs, the rest had no changes. There was a significant difference between the effects of erythromycin on GSNs and on NGSNs ( χ 2=8.34 ,P <0.05). Conclusion Erythromycin , a motilin receptor agonist, could stimulate the GSNs in LHA, which may contribute to the effect of central motilin on feeding behavior.
