茶多酚对肺癌SPC-A1细胞体外增殖及凋亡的影响 被引量:1

摘要 ①目的 探讨茶多酚对体外人肺癌SPC A1细胞增殖及凋亡的作用。②方法 体外培养人肺癌SPC A1细胞 ,以 5 0~ 30 0mg/L茶多酚作用 1 2、2 4及 36h后 ,用MTT法测定细胞的增殖活性 ,用琼脂糖凝胶电泳、透射电镜检测细胞凋亡情况。③结果 茶多酚可抑制肺癌细胞的增殖活性 ,其抑制作用随时间的延长而增强 ,以 36h抑制作用最强。在 5 0~ 1 5 0mg/L浓度范围内 ,抑制作用随浓度增高而增强 ;当浓度 >1 5 0mg/L时 ,抑制作用随浓度增高而减弱。琼脂糖凝胶电泳出现一特征性的梯状条带。透射电镜观察可见线粒体空泡化、核凝聚、核固缩等凋亡的形态学改变。④结论 茶多酚可抑制人肺癌SPC A1细胞增殖 。 Objective To study the effects of tea polyphenols(TP) on lung cancer cell line SPC A1 proliferation and apoptosis in vitro. Methods SPC A1 cell line was treated with TP at 50-300 mg/L. Cell proliferation was evaluated by MTT, and apoptosis by gel electrophoresis and electric microscope in vitro. Results The proliferation of SPC A1 was significantly inhibited by TP and the inhibition rate was time dependent (12-36 h) and concentration dependent (50-150 mg/L). Its inhibitory effect decreased when the concentration exceeded 150mg/L. “DNA ladder” was viewed on agarose gel. Typical nuclear condensation was observed by tranmissional electron microscope. Conclusion TP can inhibit the proliferation of SPC A1 in vitro, probably through inducing cancer cell apoptosis.
