氨溴索对慢性阻塞性肺疾病大鼠模型肺的保护作用 被引量:10

PROTECTIVE EFFECT OF AMBROXOL ON CHRONIC OBSTRUCTIVE PULMONARY DISEASE MODEL IN RATS

摘要 ①目的 探讨氨溴索对肿瘤坏死因子α(TNF α)在慢性阻塞性肺疾病 (COPD)大鼠模型肺内表达与分布的影响以及肺泡Ⅱ型上皮细胞超微结构的影响。②方法 实验大鼠随机分为 3组 :健康对照组 (n =8) ,COPD模型组 (n =1 2 ) ,氨溴索干预组 (n =1 2 )。采用熏吸香烟并气管内注入脂多糖法建立大鼠COPD模型 ,氨溴索干预组于COPD模型开始建立时腹腔注射氨溴索溶液 4 0mg/ (kg·d) ,连续 4 0d。观察各组气道炎症的病理特点、超微结构特点、血气指标、肺泡平均内衬间隔、平均肺泡数。应用原位杂交法检测TNF α基因表达的相对含量。③结果 所建COPD模型的病理以及病理生理学改变符合人类COPD的特点。氨溴索干预组肺泡平均内衬间隔、平均肺泡数、TNF α表达强度与健康对照组、COPD模型组比较差异有显著性 (F =3.2 4~ 1 1 .1 1 ,q =4 .74~ 36 .2 2 ,P <0 .0 1 ) ;动脉血氧分压、二氧化碳分压差异亦有显著性 (F =3.30、8.79,q =3.5 0~ 8.6 9,P <0 .0 5 )。 Objective To evaluate the expression and distribution of tumor necrosis factor α(TNF α) in the lung tissue of chronic obstructive pulmonary disease(COPD) model in rats. Methods Wistar rats were randomly divided into three groups: normal control group ( n =8);COPD model group (n =12) and ambroxol treated group ( n =12). The COPD model was established by intratracheal instillation of lipopolysaccharide and exposed to cigarette smoking. The treated group was given ambroxol,40mg/kg, daily, for 40 days, since the first day of the model completed. Pathologic changes of the lung tissue and blood gas were evaluated. MLI ,MAN were measured by computer image analyzer . The expression of TNF α was determined by in situ hybridization. Results Compared with the normal control, the model group presented significant differences in PCO 2, PO 2, MLI and MAN [( 7.0± 0.5)kPa vs(5.5±0.5)kPa,(10.7±1.1)kPa vs(13.1±0.4)kPa, (92±10)μm vs(52±3)μm,(268±15)/mm 2 vs(410±27)/mm 2] ( P <0.01).The expression of TNF α markedly elevated [(0.318±0.031)vs(0.197±0.036)]( P <0.01) . In comparison with the model group, blood gas analysis in the treated group improved [PCO 2(6.4±0.4 )kPa vs(7.0±0.5)kPa, PO 2( 1.9± 1.0)kPa vs(10.7±1.1)kPa]( P <0.05). The expression of TNF α decreased [ (0.282±0.043) vs(0.318±0.031)]( P <0.05) and MLI, MAN were significantly different [(83±8)μm vs(92±10)μm,(291±10)/mm 2 vs(268±15)/mm 2 ]( P <0.01). Conclusion Long term use of Ambroxol shows great inhibition in airway inflammation in COPD model of rats.
