摘要
本文应用Fura-2荧光染料技术和测定细胞存活率的方法研见了晶态S_iO_2的细胞毒性与C_a^(2+)的关系。结果表明:在含C_a^(2+)介质中,晶态SiO_2的毒性作用,使细胞内游离C_a^(2+)浓度(〔C_a^(2+)〕_i)升高。SiO_2粉尘浓度越大,或作用时间越长,〔C_a^(2+)〕_i升高越大。在无C_a^(2+)介质中,未观察到〔C_a^(2+)〕_i的明显变化。〔C_a^(2+)〕_i的升高与细胞存活率降低是相一致的。
The paper with fluorescence dye Fura-2/AM and measurement of viability,studies the relationship between eapeium ion and cytotoxicity of crystalline SiO2. The reslts are mainly as follows; In the Ca2+-medium, the poisonous effect of crystalline SiO2 toalveolar macrophage leads to the increase ofi the cytoplasmic free Cl2+ concentration ( [Ca2+]i). The higher the concentration of SiO2 dust is, or the longer the time of action is, the greater the increase of [Ca2+] i is. But in the absence of Ca2+, we did not find the increase of [Ca2+] i.The increase of [Ca2+] i accords with the decrease of cell's viability.
