摘要
以台盼兰排染法及中性红活染比色法测定雷公藤内酯对 L_(1210),HeLa 等癌细胞的体外杀伤作用,以[~3H]TdR,[~3H]UR 参入、荧光染色等方法观察药物对这些细胞株核酸合成的影响,发现该药对癌细胞的杀伤作用与其抑制核酸合成有关。
The cytocidal action of triptolide on L_(1210)cells and HeLa cells was determined by trypan blue dye exclusion and vital staining microplate cell culture.The IC_(50)the drug on L_(1210)and HeLa cells were 10,96ng/ml respectively.The incorporation of[~3H]thymidine,[~3H]uridine into DNA,RNA in L_(1210) and HeLa cells were inhibited by triptolide.The inhibition depended on dosage and time,and it seemed to be more remarkable in DNA synthesis than in RNA synthesis.It is suggested that the inhibition of DNA synthesis is one of the mechanisms of its cytocidal activity.
