摘要
降钙素基因相关肽家族是一类多功能的激素家族 ,参与人体的多种生物学功能 ,与多种疾病有关。降钙素基因相关肽受体包括降钙素受体 (CTR)和降钙素受体样受体 (CRLR) ,CTR可以独自与降钙素结合 ,而CRLR必须与一组称作受体活性修饰蛋白 (RAMPs)的蛋白质共同作用才能发挥生物学功能。综述CTR的研究概况及CRLR与RAMPs相互作用的机制和表达调控 ,以期为人们设计新型药物提供参考。
The calcitonin gene related peptides family is a unique family of peptide hormones important for homeostasis in diverse tissues.They consist of calcitonin,calcitonin gene related peptide(CGRP),adrenomedullin(ADM) and amylin,recently adding calcitonin receptor stimulating peptide(CRSP,from the acid extract of the porcine brain) and intermedin.Receptors of CGRP contain calcitonin receptor (CTR) and calcitonin receptor like receptor(CRLR).Unlike the interaction between calcitonin and CTR,CRLR require one of three accessory receptor activity modifying proteins (RAMPs) for ligand recognition.CRLR combining with RAMP1 function as a CGRP receptor.RAMP2 and RAMP3 enable the same CRLR to behave as an adrenomedullin receptor.Although the CTR does not require RAMP to bind and respond to calcitonin,it can associate with the RAMPs,resulting in a series of receptors that typically have high affinity for amylin and varied affinity for CGRP.Here we reviewed the progress of CTR,described the biological potency,action mechanism and transcription of RAMPs and CRLR,which will bring new ideas to develop new drugs.;
