携带肝细胞生长因子基因的重组腺病毒对犬心肌缺血的基因治疗 被引量:5

摘要 为了进一步研究重组腺病毒Ad-HGF对心肌缺血的治疗效果及其安全性,在体外实验和大鼠实验的基础上制作了犬心肌缺血的模型,从3个方面评价Ad-HGF对心肌缺血的治疗效果.首先,通过冠状动脉造影观察侧支循环形成的情况,发现治疗组动物的侧支循环比对照组丰富;然后,对心脏切片进行TTC染色,并通过图像分析确定缺血区面积,结果显示治疗组动物的缺血区明显小于对照组;最后,从心肌中回收彩色微球计算局部心肌血流量,结果显示治疗组动物的局部心肌血流量基本恢复到了结扎前的水平.另外,在长期毒性研究中,用中和方法检测了腺病毒抗体产生的情况,发现腺病毒抗体在第4次注射后产生,达到高峰后缓慢下降,于停药12周后消失.总之,Ad-HGF对心肌缺血的治疗是有效的,同时也会诱导机体产生抗体中和体内的腺病毒,这可能是腺病毒载体的清除机制.
