抑肽酶和肿瘤坏死因子-α抗体对大鼠肝缺血再灌注损伤的保护作用 被引量:5

摘要 目的 :探讨抑肽酶和肿瘤坏死因子 α(TNF α)抗体对大鼠肝缺血 再灌注损伤的保护作用。 方法 :雄性SD大鼠 1 2 0只 ,分为正常对照、缺血 再灌注 ,TNF α抗体处理和抑肽酶 +TNF α抗体处理四组 ,在肝缺血 6 0min及再灌注 1、3、6和 1 2h后 ,检测血清丙氨酸氨基转移酶 (ALT)、丙二醛 (MDA)及肝组织病理学变化。 结果 :肝缺血 再灌注后 ,肝瘀血明显 ,血浆ALT和MDA含量均显著增高 ;肝缺血 再灌注用抑肽酶 +TNF α抗体处理后 ,血清ALT和MDA含量明显降低 ,肝组织瘀血减轻。 结论 :抑肽酶联合TNF α抗体可以抑制大鼠肝缺血 再灌注所致的炎症反应 ,对肝缺血 Objective:To study the protective effects of aprotinin and tumor necrosis factorα(TNF α) antibody on liver ischemia reperfusion injury in rats. Methods:One hundred and twenty male SD rats were randomly divided into four groups: the normal control group, ischemia group, TNF αantibody group and aprotinin + TNF α antibody group. The animals were killed at 1, 3, 6, 12 hours after 60 minutes ischemia and followed reperfusion. Serum alanine aminotransferase(ALT), malondialdehyde(MDA) and liver histopathology were observed. Results: After ischemia reperfusion, the serum ALT and MDA were remarkedly increased , and the hepatic congestion was obviously. Aprotinin and TNF α antibody could decrease serum ALT and MDA levels significantly, and relieve hepatic congestion. Conclusion:Aprotinin and TNF α antibody could suppress the inflammatory reaction induced by hepatic ischemia reperfusion, and has protective effects on rat hepatic ischemia reperfusion injury.
