细菌脂多糖诱导人胃上皮癌细胞株BGC-823环氧化酶-2的合成 被引量:1

摘要 目的 :研究细菌脂多糖 (lipopolysaccharide,LPS)对人胃上皮细胞环氧化酶 1和环氧化酶 2合成的影响。方法 :实验选用LPS(E .Coli 0 111∶B4 )和人胃上皮癌细胞株BGC - 82 3。环氧化酶的合成用免疫蛋白印迹法(WesternBlotting)测定。 结果 :LPS对BGC 82 3细胞环氧化酶 2的合成有明显的诱导作用。BGC - 82 3细胞环氧化酶 - 2的含量在LPS作用 1h后明显增加 ,并持续至少 6h。结论 :E .Coli的LPS可诱导BGC - 82 3细胞COX- 2的合成。 Objective To investigate the effect of lipopolysaccharide (LPS) on the synthenase of cyclooxygenase-1 and cyclooxygenase-2 in human gastric epithelial cell line, BGC-823. Methods LPS (E. Coli 0111:B4) and BGC-823 were used in our study. The synthesis of cyclooxygenase was examined by Western blot. Results The results of Western blot showed that the content of cyclooxygenase-2 increased obviously after the cells incubated with lipopoly saccharide a hour and it could last for six hours at least, the quantity of COX-1 did not change during this period. Conclusions E. Coli LPS would be effective stimuli of the synthesis of COX-2 in gastric carcinoma epithelial cell line.
