摘要
从水囊引产的5^+月孕龄胎儿肝脏中提取RNA,分离含poly(A)尾的信使RNA[poly(A)^+mRNA],合成双链cDNA,定向插入λgtll表达载体,转染Y1090受体菌,构建了人胎肝cDNA文库。该文库有1.35×10~6个噬菌体,重组率为87.5％,可用寡核苷酸探针和单克隆抗体两种方法筛选目的基因。
Total RNA and poly(A)+ mRNA was extracted from liver tissue derived from a fetus of 5+ months gestation. ds-cDNA was synthesised and inserted into expressive vector-λgt11 directively, which infected the host strain-Y1090 then. The cDNA library of human fetal was constructed, which contained 1.35 × 106 plaques, and whose recombination rate was 87.5%. It can be screened both by oligonucleotide probe and by monoclonal antibody for the interested gene.
