血糖安对2型糖尿病大鼠小肠上皮GLUT2 mRNA表达的影响

摘要 目的 观察葡萄糖转运蛋白 2 (GLUT2 )在 2型糖尿病大鼠小肠上皮细胞的表达与分布 ,以及血糖安对其mRNA表达的影响 ,以进一步探讨血糖安抗高血糖的作用机制。方法 用高脂饲料灌胃正常大鼠 ,引起肥胖 ,然后给大鼠腹腔注射四氧嘧啶 ( 12 0mg/kg) ,连续造模两天后稳定 72h ,筛选空腹血糖值大于 16 .7mmol/L大鼠为 2型糖尿病模型组 ,根据组间差异小于 1.11mmol/L将其分组。连续给药 1周后 ,尾静脉采血测定空腹血糖值及糖耐量 ,并从大鼠小肠取材 ,应用原位杂交技术检测GLUT2的mRNA表达。结果 GLUT2在糖尿病大鼠小肠上皮细胞中表达增多 ;血糖安可减少 2型糖尿病大鼠小肠上皮细胞中GLUT2mRNA的表达。 Objective To observe the expression and distribution of GLUT2 and the effect of Xuetangan(XTA) on its mRNA expression and so as to discuss its further role in the mechanism of type 2 diabetes.Methods Normal rats were fed with highly fatted food in order to develop obesity.Then alloxan was given 120mg/kg twice a day intraperitoneally and after 72h those with plasma glucose over 16.7 mmol/L were assigned to type 2 diabetes model group.XTA was injected intraperitoneally for one week and then samples were taken from upper intestine to test the GLUT2 mRNA expression with in situ hybridization assay.Results GLUT2 increased and XTA could decrease GLUT2 mRNA expression in epithelial cells of type 2 diabetes.Conclusion XTA can reduce high level plasma glucose by increase GLUT2 mRNA expression in epithelial cells of rat intestine.
