亚低温对脑缺血再灌注后脑皮质梗塞灶及MCP-1含量的影响

Effect of mild hypothermia on the cortical infarct size and the level of MCP-1 in the ischemic core of rat cortex after transient focal cerebral ischemia

摘要 目的 观察缺血过程中亚低温对大鼠脑缺血再灌注后脑皮质梗塞灶及缺血核心区皮质内单核细胞趋化蛋白 (MCP) 1含量的影响。方法  80只雄性Wistar大鼠随机分为常温组 ( 37℃ )和亚低温组 ( 32~ 33℃ ) ,对大鼠进行神经病学评分 ,2 ,3,5三苯基四氮唑 (TTC)染色法测定缺血 2h再灌注不同时间 ( 0、6、16、2 4和 4 8h)脑梗塞灶的大小 ,用ELISA法测定缺血核心区脑皮质内MCP 1含量的变化。结果 脑缺血 2h再灌注 6h脑皮质出现明显的梗塞灶 ,亚低温能显著减小再灌注后不同时间脑梗塞灶的面积和神经病学评分 (P <0 .0 5 )。缺血核心区脑皮质内MCP 1含量于再灌注后 6h开始升高 ,至 4 8h逐渐达到高峰。亚低温能显著降低再灌注后 6h及 4 8h缺血核心区脑皮质内MCP 1的含量(P <0 .0 5 )。结论 抑制缺血再灌注后脑皮质内MCP 1的分泌 ,可能是亚低温发挥脑保护作用的重要途径之一。 Objective To examine the effect of intraischemic mild hypothermia on the cortical infarct size and the protein level of monocyte chemoattractant protein (MCP) 1 in the ischemic core of rat cortex after transient focal cerebral ischemia.Methods Eighty male Wistar rats were randomly divided into either normothermic (37℃) or mild hypothermic (32~33℃) groups,and subjected to the right middle cerebral artery occlusion for two hours,then killed at 0,6,16,24 and 48 hours after reperfusion.All the rats were given neurological evaluation.2,3,5 triphenyltetrazolium chloride (TTC) staining was used to show the cortical infarct sizes.The protein levels of MCP 1 were measured by ELISA.Results The hypothermia could significantly improve the rat neurological scale(P<0.05).The cores of cortical infarct were showed by TTC staining six hours after reperfusion and the hypothermia reduced the cortical infarct sizes significantly(P<0.05).The protein level of MCP 1 was markedly increased at 6 hours in the ischemic core of cerebral cortex,and peaked at 48h after reperfusion.The protein level of MCP 1 in the mild hypothermic groups was significantly lower than that in the normothermic groups at 6 and 48h after reperfusion(P<0.05).Conclusion These findings suggest that reducing the protein level of MCP 1 in the cerebral cortex after ischemia/reperfusion may be one of the important ways for the neuroprotective effects of mild hypothermia.
