肌萎缩侧索硬化的基因治疗

摘要 肌萎缩侧索硬化(ALS)是一种进行性神经变性疾病,由于上、下运动神经元变性导致延髓、四肢、躯干等部位肌肉逐渐无力和萎缩,确诊后3~5年患者多因呼吸肌麻痹而死亡,目前尚无有效的治疗方法。利鲁唑(riluzole)为谷氨酸拮抗剂,可使ALS患者的生存期平均延长3个月以上,是目前唯一被美国FDA批准的临床药物。其他的治疗措施还包括维生素、神经营养因子及干细胞移植等,但都没有确切效果。基因治疗为治疗ALS提供新的治疗策略。
