绞股蓝皂苷对2型糖尿病并NAFLD大鼠肝组织TNF-α mRNA表达和血硫化氢的影响 被引量:11

Effects of Gypenoside on TNF-α mRNA Expression and Plasma H 2 S in Rats with Type 2 Diabetes Mellitus and Nonalcohol Fatty Liver Disease

摘要 目的:观察绞股蓝皂苷(gypenoside,GPS)对2型糖尿病(Type 2 diabetes mellitus,T2DM)并非酒精性脂肪性肝病(nonalcohol fatty liver disease,NAFLD)大鼠肝组织肿瘤坏死因子-α(tumor necrosis factor alpha,TNF-α)mRNA和血硫化氢(hydrogen sulfide,H2S)的影响。方法:65只SPF级雄性SD大鼠随机分为空白对照组(N组),NAFLD模型组(NM组),T2DM并NAFLD模型组(模型组)。T2DM并NAFLD模型大鼠分别给予GPS低、高剂量(0.5、1.0 g·kg-1·d-1)干预(JL组、JH组),模型组以纯净水灌胃作为对照组(M组)。检测血糖(blood sugar,BS)、甘油三酯(triglycerides,TG)、总胆固醇(total cholesterol,TC)、H2S变化,定量PCR技术检测肝组织TNF-αmRNA。结果:⑴血浆H2S浓度变化:JH组、JL组显著高于M组,JH组显著高于JL组(P均<0.01);⑵血糖和TG水平:JH组、JL组显著低于M组(P均<0.01),JH组低于JL组(P<0.01,P<0.05);⑶肝组织TNF-αmRNA PCR检测:JH组、JL组显著高于M组(P<0.01),JH组高于JL组(P<0.05)。结论:GPS能降低T2DM并NAFLD大鼠的BS、TG、TC,机理与下调肝组织TNF-αmRNA表达,升高血浆H2S浓度有关。 Objective To observe the effects of gypenoside on expression of TNF-α mRNA and plasma H 2 S in rat with type 2 diabetes mellitus and nonalcoholic fatty liver disease.Methods Sixty-five SPF male SD rats were randomly divided into blank control group(group N),NAFLD model group(group NM),NAFLD and T2DM model groups(group M,JL,JH).The rats in group JL and group JH were given gypenoside with doses of 0.5 and 1.0 g·kg-1·d-1,respectively.The rats in group M were given purified water as control.The plasma levels of TG,TC,BS and H 2 S in rats were determined,the expression of TNF-α mRNA was determined by quantitative PCR technique.Results ⑴H 2 S levels of rats in group JH and JL were obviously higher than that of group M,and group JH was obviously higher than that of group JL(both P < 0.01);⑵BS and TG levels of rats in group JH and JL were obviously lower than that of group M(both P < 0.01),and group JH was significantly lower than that of group JL(P < 0.01,P < 0.05);⑶Expression of TNF-α mRNA in liver tissue of group JH and JL were obviously lower than that of group M(P < 0.01),and group JH was significantly lower than that of group JL(P < 0.05).Conclusion Gypenoside could decrease the blood sugar,triglycerides and total cholesterol in rats with type 2 diabetes mellitus and nonalcoholic fatty liver disease,which may be related with its down-regulation of TNF-α mRNA expression and up-regulation of hydrogen sulfide.
