摘要
目的:利用真核细胞表达载体pcDNA3.1(+)构建人血管内皮细胞生长因子融合表达质粒pcD-NA3-VEGF。方法:采用PCR技术和DNA重组技术。结果:将编码人VEGF基因的cDNA序列克隆至表达载体pcDNA3.1(+)上,DNA测序实验结果证明cDNA序列正确,cDNA序列是由Kozak序列,翻译起始密码ATG,信号序列,编码序列和终止密码子等部分组成。结论:通过研究获得了VEGF细胞因子的融合表达质粒pcDNA3-VEGF,为进一步研究VEGF表达载体进行转基因治疗奠定基础。
Objective: To construct the plasmid of pcDNA3-VEGF for fusional expression of human vascular endothelial growth factor with eukaryotic expression vector pcDNA3.1(+). Methods: PCR and DNA recombination methods in vitro were used. Results:A gene fragment encoding VEGF was cloned into the fusional expression vector pcDNA3.1(+). DNA sequence analysis indicated that it was correct. The sequence of VEGF cDNA was composed of Kozak sequence, translation original codon, ATG, signal sequence,coding sequence and terminating codon. Conclusion: A fusional expression plasmid pcDNA3 VEGF was successful constructed. From the basis of the work, further study of transgenic therapy with VEGF could be carried out.
