摘要
目的探讨氨茶碱/壳聚糖/β-环糊精微球肺部缓释微球的呼吸道粘膜毒性。方法通过测定氨茶碱/壳聚糖/β-环糊精微球肺部给药后肺泡灌洗液乳酸脱氢酶活性和总蛋白及纤毛毒性实验,对其安全性进行评价。结果氨茶碱/壳聚糖/β-环糊精微球的纤毛持续运动时间为512.33 min,纤毛运动频率为96.0%,表明其有良好的生理适应性;氨茶碱/壳聚糖/β-环糊精微球气管给药后总蛋白含量增加,乳酸脱氢酶(LDH)活性与正常组和阴性对照组差异不明显。结论氨茶碱/壳聚糖/β-环糊精微球对呼吸道粘膜无明显毒性,可作为肺部给药载体。
Objective To explore mucociliotoxicity of theophylline/chitosan/β-cyclodextrin microspheres as pulmonary sustained drug delivery carriers.Methods An acute cell toxicity experiment(LDH and protein in BALF) and a mucociliotoxicity experiment were carried out.Results Ciliary movement time of MS B microspheres was 512.33 minutes and ciliary beat frequency was 96.0%.The total contents of protein and LDH in BALF of the pulmonary delivery group were not significantly different from those of the control group.Concl...
