SPK/S1P信号途径对人肝癌细胞VEGFmRNA表达的作用 被引量:7

摘要 目的探讨鞘氨醇激酶/1-磷酸鞘氨醇信号途径(SPK/S1P)在基因水平对人肝癌细胞分泌血管内皮生长因子(VEGF)的调节作用。方法体外培养人肝癌细胞HepG2,利用荧光定量PCR法检测鞘氨醇激酶的特异性抑制剂N,N′-二甲基鞘氨醇(DMS)作用后肝癌细胞VEGFmRNA的表达。结果不同浓度DMS处理后的HepG2细胞中VEGF基因表达较对照组明显降低,与对照组比较差异有统计学意义(P<0.01)。结论DMS可通过阻断SPK/S1P信号途径在基因水平抑制肝癌细胞VEGFmRNA,说明人肝癌细胞VEGFmRNA表达受SPK/S1P信号途径调控。 Objective To investigate the role of the SPK/S1P(sphingosine kinase/sphingosine-1-phospate)signal on gene level in regulating expression of vascular endothelial growth factor(VEGF) in human hepatoma cells.Methods The human hepatoma cell line HepG2 was cultured in vitro.After treatment with a concentration of 520μmol/L DMS(dimethyl sphingosine) in culture media,VEGFmRNA expression was determined by RT-PCR.Results Expression of VEGFmRNA was obviously inhibited by DMS in a dose-dependent manner.Conclusion DMS ...
