摘要
研究了用ALuANcE自动分析仪测定烟草中的果胶的方法.样品经80%乙醇回流抽提后,在55～60℃和pH为4.0的磷酸盐缓冲溶液中用果胶酶酶解,用流动分析仪测定酶解液中的果胶含量.与传统的重量法相比,该法的回收率为97～104%,RsD为0.58～1.07%,且操作简便快速,准确度高,精密度好,便于批量样品的处理,为准确、快速地测定烟草中的果胶提供了一种便利的方法.
The determination of pectin in tobacco with ALLANCE automatic analyzer was studied.The tobacco sample was hydrolyzed with pectase under the temperature 55～60℃ in phosphate buffer solution(pH=4.0) after ethanol extraction.Then the pectin content(expressed by galacturonic acid) in the enzymolysis fluid was determined with ALLANCE automatic analyzer.The recovery rate of this method is in the range of 97～104%,and the relative standard deviation 0.58～1.07 %.Compared with traditional gravimetric analysis,this method is simple and quick in operation,accurate and précis in results convenient for batch sample test,and is suitable for the determination of pectin in tobacco.
