摘要
目的 :建立一种以不被钙离子干扰、2 - (8 -羟基喹啉 - 5 -磺酸 - 7 -偶氮 ) -变色酸 (8Q5 SAC)显色、特定波长比色的血清镁测定法。 方法 :用 8Q5 SAC作显色剂 ,在 p H10 .8、含聚乙烯吡咯烷酮 (PVP)的甘氨酸 - Na OH缓冲介质中 ,以 5 70 nm特定波长比色测定血清镁 ,并对反应条件、方法性能进行系统研究。 结果 :该法线性 0~ 2 m mol/L ,平均回收率为 99.2 % ,CV 2 .0 2 %~ 3.16 %。与 Calm agite法对照 ,r=0 .995 ,P>0 .5 ;与二甲苯基氮紫 (キシリルアゾバイオレツト I)法对照 ,r=0 .992 ,P>0 .5 ;测试灵敏度是 Calm agite法的 2倍 ,与二甲苯基偶氮紫法基本一致。 结论 :该法利用 8Q5 SAC-镁的特异吸收波长测定血清镁 ,不受钙离子干扰 ,方法简便快速 ,灵敏度高 ,适用于血清镁的常规手工或自动化分析。
Objectives:To develop a method for the determination of serum magnesium with 2 (8 quinolinol 5 sulfo 7 azo) chromotropic acid (8Q5SAC) at specific wavelength without covering calcium. Methods:, Magnesium was determined in a pH = 10.8 glycine NaOH buffer solution containing polyvinylpyrrolidone (PVP) by spectrophotometry at 570nm, 8Q5SAC was used as a color reagent. Systematic studies of the reaction conditions and properties of the method were also made. Results: Linearity was 0 2(mmol/L).The average rate of recovery was 99.2% , CV 2.02%~3.16%. Comparing with Calmagites method, r =0.995, P >0.5,and with xylyl azo violet method (キシリルアゾバイ? 欹磨菼), r =0.992, P >0.5. The method was 2 times sensitive than Calmagites method and similar to xylyl azo violet method. Conclusions:The determination method was simple, sensitive, rapid and not interfered by calcium, and suitable for routine analysis of serum magnesium manually or automatically.
