摘要
目的分析在不同溶液体系中,影响国产光敏剂血卟啉单甲醚(HMME)光漂白速率的主要因素。方法首先从理论上,根据光漂白速率方程分析了可能影响光漂白速率的主要因素。然后测定不同溶液中不同浓度HMME的光漂白(充氧),通过绘制光漂白曲线(HMME浓度-照光时间)比较HMME漂白速率。结合理论推导结果与实验结果分析各因素对漂白速率的影响。结果HMME的光漂白速率在白蛋白悬液中快于DMSO中,DMSO中快于细胞悬液和PBS中,后两种溶液中HMME的光漂白速率相近。HMME对532nm光的摩尔消光系数:εDMSO>εcells-suspension>εalbumin-buffer>εPBS。结论光敏剂溶液条件会影响光敏剂光漂白速率。其中摩尔消光系数、光敏剂浓度、溶液微环境对光漂白速率影响较大。摩尔消光系数增加HMME光漂白速率,蛋白、DMSO的存在可以加速HMME的光漂白。
Objective To analyze the principal impact factors that can affect the photobleaching rate of HMME in different solutions. Methods Firstly, the principal impact factors that might affect the photobleaching process according to a theoretic photobleaching rate equation were deduced. Then the photobleaching data of HMME were calculated by measuring the absorbance values at different time before and after laser irradiation,and the photobleaching rates of HMME were compared by drawing photobleaching curves (HMME concentration-irradiation time). The main factors that could affect the photobleaching rate of HMME were analyzed by combining the theoretic deduction and experimental results. Results Photobleaching rates of HMME were different in different solutions:albumin solution > DMSO > cells suspension ≈ PBS. The molar extinction coefficients of HMME at 532nm laser irradiation in different solutions were: ε_ DMSO>ε_ cells-suspension>ε_ albumin-buffer >ε_ PBS. Conclusions Microenvironment of solutions can affect the photobleaching rates. The molar extinction coefficients and concentrations of photosensitizer are the principal impact factors. The photobleaching speeds up with the molar extinction coefficient of photosensitizer increased. Both albumin and DMSO can accelerate the photobleaching of HMME.
