摘要
目的 探讨血管内皮生长因子(VEGF)和色素上皮衍生因子(PEDF)在氪激光诱导BN大鼠脉络膜新生血管(CNV)形成过程中的表达及其意义.方法 BN大鼠30只,随机分成5组,每组6只,单眼视网膜氪激光光凝,诱导脉络膜新生血管形成.分别在光凝后3 d,1、2、3和4周摘除眼球,应用免疫组织化学技术对光凝区进行因子Ⅷ相关抗原(FⅧR:Ag)的检测以观测CNV形成,并检测CNV形成过程中VEGF、PEDF蛋白的表达,分析其变化趋势.结果 FⅧR:Ag免疫组织化学检查结果显示,光凝后1周开始形成CNV,3~4周达到高峰.正常BN大鼠视网膜中,VEGF在视网膜神经节细胞层、内核层、色素上皮层、视网膜和脉络膜血管内皮细胞表达;PEDF在神经节细胞、内核层、色素上皮层表达.视网膜光凝后,除上述部位外,VEGF和PEDF均在外核层和脉络膜的光凝损伤区阳性表达.光凝后3 d至4周,光斑区内FⅧR:Ag、VEGF阳性染色密度逐渐增加(P<0.05),PEDF阳性染色密度逐渐下降(P<0.05);FⅧR:Ag与VEGF阳性染色密度正相关(r=0.89,P<0.05),与PEDF阳性染色密度负相关(r=-0.84,P<0.05).结论 CNV形成与VEGF表达正相关,与PEDF表达负相关,VEGF和PEDF表达失衡可能是CNV形成和增殖的主要原因之一.
Objective To study the expression of vessel endothelial growth factor (VEGF) and pigment epithelium-derived factor (PEDF) in choroidal neovascularization (CNV),and to investigate their expression and significance on CNV induced by krypton laser in BN rats. Methods Five groups of 30 BN rats were photocoagulated by krypton laser in single eye.Three days, 1 week, 2 weeks, 3 weeks and 4 weeks after photocoagulation, the expression of VEGF, PEDF and factor Ⅷ related antigen (FⅧR ∶Ag) was observed by immunohistochemical method. Results FⅧR ∶Ag positive staining cells formed vessels in the lesion 1 week after photocoagulation;the expression of FⅧR ∶Ag gradually increased till 3-4 weeks. VEGF expression was observed in the ganglion cells, inner nuclear layer, retinal pigment epithelial cells,and the vascular endothelial cells in normal retina and choroid of BN rat. VEGF expressed in the ganglion cells, inner nuclear layer, the vascular endothelial cells,and lesion of outer nuclear layer and choroid after photocoagulation. PEDF expressed in the ganglion cells,inner nuclear layer,and retinal pigment epithelial cells in normal BN retina. After photocoagulation, the expression of PEDF was observed in the ganglion cells, inner nuclear layer, retinal pigment epithelial cells, and the lesions of outer nuclear layer and choroid. From 3 days to 4 weeks, the expression density of FⅧR ∶Ag and VEGF in the lesions gradually increased(P<0.05), however, the expression density of PEDF decreased (P<0.05).The expression of FⅧR ∶Ag was positively correlated with that of VEGF(r=0.89,P<0.05)and inversely correlated with that of PEDF during the development of CNV(r=-0.84,P<0.05). Conclusions CNV is positive correlated with the expression of VEGF and inversely correlated with that of PEDF. The unbalanced expression between VEGF and PEDF may contribute to the main reason for the formation and development of CNV.
