莫索尼啶对阿霉素肾病大鼠保护作用的研究

摘要 目的:探讨莫索尼啶对阿霉素肾病大鼠肾间质纤维化的影响及其机制。方法:将30只健康雄性sD大鼠随机分为3组:模型组(Mod)、莫索尼啶治疗组(Mox)、健康对照组(Con)。采用尾静脉一次性注射阿霉素方法建立大鼠肾病模型,对照组尾静脉注射等量生理盐水。12周时测血浆去甲肾上腺素、血管紧张素Ⅱ水平。行HE及Masson染色,观察肾间质纤维化改变。免疫组化和原位杂交方法检测肾组织中转化生长因子β1(TGF-β1)蛋白及mRNA表达。结果:模型组与对照组比大鼠血浆NE、AngⅡ水平、肾间质纤维化面积、肾小管损害程度、肾小球体积均显著上升(P<0.01);治疗组较模型组明显下降(P<0.01)。肾组织内TGF-β1蛋白及mRNA的表达:模型组较对照组显著上升(P<0.01);治疗组较模型组显著下调(P<0.01)。结论:莫索尼啶能抑制交感神经活性,下调肾组织中TGF-β1的表达,干预肾间质纤维化进程来保护肾脏的。 Objective:To study the influence of Moxonidine on the renal interstitial fibrosis caused by Adrismycin induced in rats.Methods:Thirty healthy Sprague-Dawley rats were randomly divided into 3 groups:model group(Mod),moxonidine treatment group(Mox)and control group(Con).the rats received first-intravenous injection of adrismycin to induce rats model with renal fibrosis.The control group received first-intravenous injection of 0.9%Nacl.By the end of twelve weeks,the blood sample were collected to test Noradrenaline(NE)and AngiotensinⅡ(Ang Ⅱ).The kidney pathomorphism was investigated at the end of experiment.Immunohistochemistry staining and In situ hybridization were used for expression of TGF-β1 and TGF-β1 mRNA respectively.Resuits:Compared with the Con group,the indices NE and AngⅡ in the plasma,the area of fibrosis,the renal tubular damaged,volume of glomeruli were significantly increased(P<0.01)in the Mod group.Compared with Mod group there were significantly decreased in the treatment groups(P<0.01).In Mod group the expression of TGF-β1 was higher than that in Con group(P<0.01).Compared with the Mod group,the expression of TGF-β1 in Mox groups was decreased markedly(P<0.01).Conclusion:Moxomdme can protect the kidney by inhibiting the activity of sympathetic nerve and down-regulating significantly the expression of TGF-β1 and inhibiting tubulointerstitial fibrosis.
