摘要
目的:探究过表达miR-122a是否可提高肝癌细胞HepG2对化疗药物的敏感性,并阐明其提高化疗药物敏感性的部分机制。方法:构建过表达miR-122a的HepG2细胞系(miR-122a肿瘤细胞组),并与HepG2细胞(肿瘤细胞组)置于含不同浓度顺铂的培养液中,CKK-8法观察miR-122a上调对HepG2细胞存活率的影响,Annexin V-PE/7-AAD双染法观察miR-122a上调对HepG2细胞凋亡率的影响;Western blot检测miR-122a肿瘤细胞组和肿瘤细胞组中bcl-2和p53蛋白的表达水平。结果:低铂浓度顺铂条件下,miR-122a肿瘤细胞组细胞的存活率低于肿瘤细胞组细胞的存活率,miR-122a肿瘤细胞组24 h细胞的凋亡率高于肿瘤细胞组(P<0.05);Western blot法检测bcl-2和p53蛋白结果显示,在低浓度顺铂溶液中,miR-122a肿瘤细胞组bcl-2表达量低于肿瘤细胞组(P<0.05),miR-122a肿瘤细胞组p53表达量高于肿瘤细胞组(P<0.05)。结论:过表达miR-122a能上调p53蛋白表达、下调bcl-2蛋白表达,增加HepG2细胞对顺铂敏感性,为肝癌患者提供基因治疗和降低肿瘤细胞耐药提供了理论和实验基础。
Objective:To explore whether overexpression of miR-122 a can increase the sensitivity of HepG2 cells to chemotherapeutic drugs,and to clarify part of the mechanism for improving the sensitivity of chemotherapeutic drugs.Methods:Construct HepG2 cells overexpressing miR-122 a(miR-122 a tumor cell group),and place them with HepG2 cells(tumor cell group)in a culture medium containing different concentrations of cisplatin.CKK-8 method was used to observe the survival rate of HepG2 cells by up-regulation of miR-122 a and the Annexin V-PE/7-AAD double staining method was used to observe the effect of miR-122 a up-regulation on the apoptosis rate of HepG2 cells;Western blot was used to detect the expression levels of bcl-2 and p53 proteins in the miR-122 a tumor cell group and the tumor cell group.Results:The survival rate of cells in the miR-122 a tumor cell group under the condition of low platinum concentration of cisplatin was significantly lower than that of the tumor cell group,and the 24 h cell apoptosis rate of the miR-122 a tumor cell group was significantly higher than that of the tumor cell group(P<0.05);Western Bcl-2 and P53 genes detected by blot method showed that in the low-concentration cisplatin solution,the expression of bcl-2 in the miR-122 a tumor cell group was significantly lower than that in the tumor cell group(P<0.05),and the p53 in the miR-122 a tumor cell group The expression level was significantly higher than that of the tumor cell group(P<0.05).Conclusion:Overexpression of miR-122 a can up-regulate the expression of p53 protein and down-regulate the expression of bcl-2 protein,increase the sensitivity of HepG2 cells to cisplatin,and provide a theoretical and experimental basis for gene therapy for liver cancer patients and reduction of tumor cell resistance.
