摘要
为了开发用于污水治理的高絮凝活性菌株,拟对高岭土有44.8%絮凝率的芽孢杆菌2645进行16S rDNA鉴定,并利用单因素试验和正交试验对其培养基组分进行优化。经过16S rDNA鉴定表明,芽孢杆菌2645与Bacillus wiedmannii strain FSL W8-0169同源性最高,培养基优化单因素试验结果显示,培养基最佳碳源为玉米粉,最佳氮源为脱脂大豆粉和酵母粉,最佳无机盐为KCl;将脱脂大豆粉、酵母粉、KCl和玉米粉4因素按L_(16)(4^(5))设计正交试验,结果表明,最佳配比为脱脂大豆粉12 g/L、酵母粉5 g/L、KCl 6 g/L、玉米粉8 g/L。培养基组分优化后,芽孢杆菌2645絮凝率可以达95.3%,提高了1.13倍。
In order to develop high flocculating activity strains for wastewater treatment,16S rDNA identification of Bacillus 2645with 44.8%flocculating rate of kaolin was proposed,the medium composition was optimized by single factor test and orthogonal test.The identification of 16S rDNA showed that Bacillus 2645 had the highest homology with Bacillus wiedmannii strain FSL W8-0169.The results of single factor test of medium optimization showed that the optimum carbon source of medium was corn flour,the optimum nitrogen source was defatted soybean powder and yeast powder,and the optimum inorganic salt was KCl.The orthogonal test was designed according to L_(16)(4^(5))for defatted soybean powder,yeast powder,KCL and maize powder,the best ratio was defatted soybean powder 12 g/L,yeast powder 5 g/L,KCl 6 g/L,corn powder 8 g/L.The flocculation rate of Bacillus 2645 reached 95.3%,which was1.13 times higher than that of the control.
