四溴双酚A对斑马鱼子代幼鱼生长发育的影响及分子机制研究 被引量:1

Effects of meternal exposure to tetrabromobisphenol A on the growth and development of offspring larvae of zebrafish and its molecular mechanisms

摘要 目的探讨四溴双酚A(TBBPA)暴露后对水生生物后代生长发育的影响。方法以斑马鱼为受试生物,以不同浓度的TBBPA(0、60、600μg/L)暴露28 d后,收集子代胚胎,分析TBBPA在子代胚胎中的生物蓄积,并且评价子代幼鱼生长发育情况;采用qRT-PCR检测子代幼鱼下丘脑—垂体—甲状腺(HPT)轴和生长激素—胰岛素样生长因子(GH—IGF)轴相关基因,探究其可能的分子机制。结果TBBPA能在斑马鱼子代胚胎中蓄积,与对照组比较,600μg/L组子代幼鱼的体长显著缩短(P<0.05),且子代幼鱼的促甲状腺激素受体(tshr)、甲状腺激素运载蛋白(ttr)和甲状腺激素受体β(tr-β)、生长激素受体b(ghrb)、胰岛素样生长因子1受体a和b(igf1ra和igf1rb)、胰岛素样生长因子2a(igf2a)基因表达显著下调(P<0.05);60μg/L组子代幼鱼ghrb和igf2a基因表达显著下调(P<0.05),而脱碘酶1(dio1)显著上调(P<0.05)。结论TBBPA暴露会影响斑马鱼子代幼鱼HPT轴和GH—IGF轴相关基因的表达,影响子代幼鱼的体长发育。 Objective To understand the effects of maternal exposure to tetrabromobisphenol A on growth and development of offspring of aquatic life and its molecular mechanisms.Methods Adult zebrafish were exposed to TBBPA at different concentrations(0,60 and 600μg/L)for 28 days,and then the exposed fish were spawned in TBBPA-free water.The TBBPA contents were measured in offspring embryos and developmental endpoints were examined in larval offspring.The expressions of genes associated with hypothalamus-pituitary-thyroid(HPT)axis and growth hormone/insulin-like growth factor(GH-IGF)axis in offspring were further measured for exploring the possible mechanisms.Results TBBPA could be accumulated in offspring.Compared with the control group,the body length of larval offspring was significantly decreased following exposure of parents to TBBPA.TBBPA could be accumulated in offspring embryos and body length of offspring larvae was significantly decreased in 600μg/L TBBPA exposure group(P<0.05),which accompanied by down-regulation of expressions of thyroidstimulating hormone receptor(tshr),transthyretin(ttr),thyroid receptorβ(th-β),growth hormone receptor a(ghra),insulin-like growth factor 2 a(igf2 a),insulin-like growth factor 1 receptor a(igf1 ra)and igf1 rb(P<0.05).In 60μg/L TBBPA exposure group,the expressions of ghra and igf2 a were significantly down-regulated,while the expression of deiodinase(dio1)was significantly up-regulated collectively(P<0.05).Conclusion TBBPA can subsequently inhibit the body length of larval offspring by disrupting the expressions of genes involved in HPT axis and GH-IGF axis.
