急性百草枯染毒对大鼠学习记忆能力的影响 被引量:1

Effects of acute exposure to paraquat on learning and memory ability in rats

摘要 目的通过行为学实验探索急性百草枯染毒对大鼠学习记忆能力的影响。方法将72只SPF级成年Wistar大鼠按体重随机分为对照(生理盐水)组、25和45 mg/kg百草枯染毒组,每组24只,雌雄各半。采用一次性经口灌胃方式进行染毒。分别于染毒后第9、30、60天进行Morris水迷宫实验,Morris水迷宫实验结束第3天进行跳台实验,测试大鼠学习记忆能力。结果与对照组比较,染毒后第30天时45 mg/kg百草枯染毒组雌性大鼠定位航行实验第1、2、4天的平均逃避潜伏期及染毒后第60天时45 mg/kg百草枯染毒组雌性大鼠定位航行实验第1、4天的平均逃避潜伏期均延长;仅染毒后第30天时45 mg/kg百草枯染毒组雄性大鼠定位航行实验第1、2天的平均潜伏期延长,差异均有统计学意义(P<0.05)。染毒后第9天时,各剂量百草枯染毒组大鼠的平均潜伏期与对照组比较,差异均无统计学意义(P>0.05)。与对照组相比,染毒后第9天及60天时各剂量百草枯染毒组大鼠的穿台次数及目标象限活动时间均减少,染毒后第30天时45 mg/kg百草枯染毒组大鼠的目标象限活动时间减少,差异均有统计学意义(P<0.05);而染毒后不同时间各剂量百草枯染毒组大鼠的空间探索时间及路径比例均无明显改变。与对照组比较,仅染毒后第37天时45 mg/kg百草枯染毒组雄性大鼠训练第2天的平均逃避潜伏期缩短,差异有统计学意义(P<0.05);而染毒不同时间各剂量百草枯染毒组大鼠的错误次数均无明显改变。结论急性百草枯染毒可在一定时期内对大鼠的学习记忆能力产生不良影响。 Objective To understand the effects of acute exposure to paraquat on learning and memory ability in rats by neurobehavioral assessment.Methods A total of 72 healthy adult Wistar rats,half male and half female,were randomly divided into three dose groups,such as the control group(normal saline),25 and 45 mg/kg paraquat exposure groups,which were exposed to paraquat through intragastric administration once.Each group was then randomly divided into three sub-groups,the learning and memory ability of which were evaluated by using Morris water maze test at the 9 th,30 th and 60 th day after the acute exposure to paraquat respectively,the step-down test was carried out three day after the Morris water maze experiment.Results Compared with the control group,at the 30 th day after exposure,the average latency of escaping onto platform of place navigation test among female rats in the 45 mg/kg paraquat exposure group was prolonged at the 1 st、2 nd and 4 th training day;At the 60 th day after exposure,the average latency of escaping onto platform of female rats in the 45 mg/kg paraquat group was prolonged At the 1 st and 4 th training day;At the 30 th day after exposure,the average latency of escaping onto platform of the male rats in the 45 mg/kg paraquat exposure group was only prolonged at the first two training days,the difference was statistically significant(P<0.05);And no significant changes were seen in the average latency of escaping onto platform among three paraquat exposure groups at the 9 th day after exposure(P>0.05).Compared with the control group,at the 9 th and 60 th day after exposure,the number of crossing platform and the target quadrant activity time of spatial probe test among rats in the 25 and 45 mg/kg paraquat exposure groups decreased,and the target quadrant activity time of rats in the 45 mg/kg paraquat group decreased at the 30 th day after exposure,the difference was statistically significant(P<0.05);And no significant change was seen in space exploration time or path selection among paraquat exposure groups.Compared with the control group,the average escape latency of the step-down test among male rats in the 45 mg/kg paraquat group was shortened on the second day of training at the 37 th day after exposure with significant differences(P<0.05);However,there was no significant changes in the number of errors among rats in paraquat exposure groups at different periods after exposure.Conclusion Acute paraquat exposure may have adverse effects on learning and memory ability in rats in a certain period after exposure.
