Analysis of NA molecular characteristics of influenza A(H1N1) virus in Zhumadian,Henan from 2018 to 2019

摘要 目的了解驻马店市2018-2019年度甲型H1N1流感病毒神经氨酸酶(NA)基因进化特征及关键位点氨基酸突变和抗原变异情况。方法选取2018-2019年度驻马店市分离的6株甲型H1N1流感病毒株,对NA基因进行测序及基因序列分析。结果驻马店市甲型H1N1流行株与2008年世界卫生组织(WHO)推荐的疫苗株A/California/07/2009核苷酸和氨基酸同源性分别为96.70%~97.00%和95.30%~98.50%,与同期疫苗株A/Brisbane/02/2018核苷酸和氨基酸同源性分别为99.00%~99.40%和98.10%~99.10%。驻马店市流行株NA蛋白28处氨基酸位点发生变异,其中在药物结合位点V241I和N369K、抗体结合位点N200S、K432E和N248D、抗原区V264I、N270K、I321V和N386K氨基酸位点共同变异,分离出1株E119K。结论2018-2019年度驻马店市流感监测病例中首次发现1株E119K,该位点的变异降低了对神经氨酸酶抑制剂的敏感性,有必要对驻马店市甲型H1N1流感病毒进行加强监测。 Objective The study aimed to analyze the characteristics of neuraminidase(NA)gene evolution and mutation of antigenic epitopes of influenza A(H1 N1)virus in Zhumadian during the influenza season of 2018-2019.Methods In total,6 strains of influenza A(H1 N1)virus isolated in Zhumadian were selected for NA gene sequencing and analyzing.Results Between influenza A(H1 N1)virus strains from Zhumadian and influenza A/California/07/2009(H1 N1)vaccine strains recommended by WHO,the homologies of nucleotide and amino acid were 96.70%-97.00%and 95.30%-98.50%,respectively;the homologies of nucleotide and amino acid between these isolated strains and reference strain A/Brisbane/02/2018 were 99.00%-99.40%and98.10%-99.10%,respectively.28 obvious amino acid mutations were detected in the NA gene of Zhumadian strains including V241 I and N369 K(drug binding sites),N200 S,K432 E and N248 D(receptor binding sites),V264 I,N270 K,I321 V and N386 K(antigenic epitopes).In addition,the variation of E119 K resistant locus in NA protein was found in 1 strains.Conclusion For the first time,one influenza A(H1 N1)virus strain with E119 K mutation in NA gene was detected in influenza surveillance cases in Zhumadian from 2018 to 2019,which decreased sensitivity to neuraminidase.Continuous surveillance is necessary for control and prevention of influenza A(H1 N1).
