摘要
目的使用网络药理学研究方法构建半枝莲-半边莲药对基于多成分、多靶点、多通路作用于胃癌的机制。方法利用TCMSP、TCMID、PharmMapper数据库获取药物有效活性成分及药物对应潜在靶标,利用GeneCards、OMIM及DisGeNET数据库筛选胃癌相关靶点;利用String数据库及Cytoscape 3.7.2构建化合物-靶点网络、Venn图以及PPI网络图;利用ClueGO及CluePedia插件对筛选出的基因进行GO功能和KEGG通路富集分析。结果共筛选出半枝莲-半边莲44种化学成分,对应301个靶点,得到作用于胃癌的交集靶点157个,GO功能分析集中在对细胞死亡、凋亡的调控,对多种蛋白激酶活性的调节等方面,KEGG分析得到PI3K-Akt信号通路和MAPK信号通路等重要通路。结论该研究结果预测了半枝莲-半边莲药对治疗胃癌的可能作用靶点及通路,但尚有不足之处,需要后期实验研究验证。
Objective To predict the action targets of anti-gastric cancer active ingredients of Banzhilian(Scutellariae Barbatae Herba)-Banbianlian(Lobeliae Chinensis Herba)in order to explore the“multi-component,multi-target and multi-pathway”mechanism using network pharmacology.Methods The active ingredients of Banzhilian(Scutellariae Barbatae Herba)-Banbianlian(Lobeliae Chinensis Herba)that obtained through Traditional Chinese Medicine Systems Pharmacology Database andAnalysis Platform(TCMSP),traditional Chinese medicine integrative database for herb molecular mechanism analysis(TCMID)were used to predict the targets by the reversed pharmacophore matching method.The gastric cancer-related targets were screened out by searching databases,such as Online Mendelian Inheritance in Man(OMIM),GeneCards and DisG eNET.Then components-target network and Venn maps were constructed.The effective targets of drug-disease interaction were screened.The effective targets in STRING database and Cytoscape3.7.2 were importedto get PPI.The effective targets were analyzed byClueGO and CluePedia for gene ontology(GO)and KEGG related pathway enrichment and analysis.Results Totally 44 active components and 301 effective targets of Banzhilian(Scutellariae Barbatae Herba)-Banbianlian(Lobeliae Chinensis Herba)were screened.A total of 157 effective targets of drug-disease interaction were screened.The GO pathway enrichment analysis of the targets showed that major entries were regulation of apoptotic process,cell death and multiple protein kinase activity.KEGG pathway enrichment analysis showed several pathways such as MAPK signaling pathway and PI3 K-Akt signaling pathway.Conclusion The results of study predict the possible mechanism of Banzhilian(Scutellariae Barbatae Herba)-Banbianlian(Lobeliae Chinensis Herba)treating gastric cancerand provide a theoretical basis for the later experimental study.
