基于网络药理学探讨安宫牛黄丸加味治疗新型冠状病毒作用机制 被引量:6

Study on the Mechanism of Modified Angong Niuhuang Pill(安宫牛黄丸)in the Treatment of SARS-CoV-2 Based on Network Pharmacology

摘要 目的运用网络药理学技术探寻安宫牛黄丸加味治疗新型冠状病毒(SARS-CoV-2)的作用机制,为中药在防治新型冠状病毒方面发挥更大的临床作用。方法通过检索TCMSP数据库、Batman数据库获得安宫牛黄丸加味方药中各药物的有效活性成分及相关靶点,新型冠状病毒的疾病数据通过OMIM和GeneCards数据库获取,运用Perl制作药物-疾病韦恩图并获取交集靶点,运用Cytoscape3.7.1软件构建药物-成分-靶点-疾病相互作用网络,String数据库生成安宫牛黄丸治疗新型冠状病毒的蛋白与蛋白相互作用(PPI)网络,R×643.6.1软件分别用来进行PPI网络中关键蛋白可视化、交集靶点基因本体(GO)分类富集分析和京都基因与基因组百科全书(KEGG)通路分析。结果安宫牛黄丸加味可能通过F2、CD36、ADRB2、THBD、APOE、SERPINE1、SERPINE2、CAT、RYR1、PRKDC、PIK3CD等靶点对新型冠状病毒起到治疗作用,其过程涉及的关键蛋白包括INS、IL6、TNF、AKTI、VEGFA、MAPK3,涉及的生物过程包括蛋白酶结合、受体配体活性、细胞因子活性、磷酸酶结合、丝氨酸型内肽酶活性、细胞因子受体结合、丝氨酸肽酶活性、丝氨酸水解酶活性等,涉及的信号通路包括了HIF-1信号通路、Toll样受体信号通路、TNF信号通路、NF-κB信号通路、JAK-STAT信号通路等。结论网络药理学方法能够在一定程度上阐述安宫牛黄丸加味治疗新型冠状病毒的作用机制,显示出了安宫牛黄丸加味治疗新型冠状病毒是通过多靶点、多生物过程及多个信号通路起作用,为临床用药提供了一定的理论依据。 Objective To explore the mechanism of the treatment of SARS-Co V-2 with modified Angong Niuhuang Pill(安宫牛黄丸)by using network pharmacology technology,and to play a greater clinical role for the prevention and treatment of a new coronavirus by traditional Chinese medicine.Methods By searching the TCMSP database and the Batman database,the active ingredients and related targets of each drug in the Angong Niuhuang Pill flavored prescription were obtained.The disease data of SARS-Co V-2 were obtained through the OMIM and Gene Cards databases.Perl was used to make the drug-disease Wayne diagram and obtain the intersection target,using Cytoscape3.7.1 software to build the drug-component-target-disease interaction network,and the String database generates the protein-protein interaction(PPI)network of Angong Niuhuang Pill for the treatment of SARS-Co V-2,R×643.6.1 software was utilized to visualize key proteins in the PPI network,classify enrichment analysis of target gene ontology(GO)and pathway analysis of Kyoto Gene and Genome Encyclopedia(KEGG).Results Modified Angong Niuhuang Pill may play a therapeutic role against new coronavirus(SARS-Co V-2)through targets such as F2,CD36,ADRB2,THBD,APOE,SERPINE1,SERPINE2,CAT,RRY1,PRKDC,PIK3 CD,etc.The key proteins include INS,IL6,TNF,AKTI,VEGFA,MAPK3.The biological processes involved include protease binding,receptor ligand activity,cytokine activity,phosphatase binding,serine endopeptidase activity,cytokine receptor binding,serine peptidase activity,serine hydrolase activity,etc.The involved signaling pathways include HIF-1 signaling pathway,Toll-like receptor signaling pathway,TNF signaling pathway,NF-κB signaling pathway,JAK-STAT signaling pathway,etc.Conclusion The network pharmacology method can explain the mechanism of the treatment of the new coronavirus with the modified Angong Niuhuang Pill to a certain extent,which shows that the treatment of the SARS-Co V-2 with the modified Angong Niuhuang Pill is through multiple targets,multiple biological processes and multiple signaling pathways to play a role,providing a certain theoretical basis for clinical medication.
