前列腺癌组织c-Met、MMP-2 mRNA表达变化及临床病理意义

Changes of mRNA expression of c-Met and MMP-2 in prostate cancer and their clinicopathological significance

摘要 【目的】观察前列腺癌(prostate cancer,PCa)组织中间质表皮转化因子(c-mesenchymal-epithelial transition factor,cMet)、基质金属蛋白酶2(matrix metallo proteinases-2,MMP-2)mRNA表达变化,并探讨其临床病理意义。【方法】收集经手术切除且病理证实的54例PCa组织和24例良性前列腺增生组织(benign prostatic hyperplasia,BPH),采用PCR法检测组织中c-Met、MMP-2 mRNA表达,观察c-Met、MMP-2 mRNA表达在PCa和BPH组织中的差异,及其与PCa的病理分级、临床分期及侵袭转移的关系,分析PCa与BPH组织中c-Met、MMP-2 mRNA表达的相关性;用ELISA法检测PCa和BPH患者血清肝细胞生长因子(hepatocyte growth factor,HGF)含量,比较两组HGF含量差异,并分析PCa患者血清HGF含量与组织c-Met、MMP-2 mRNA表达的相关性。【结果】C-Met、MMP-2 mRNA在PCa组织中表达量显著高于BPH组织(P<0.05);PCa组织中c-Met与MMP-2 mRNA表达呈正相关关系(r=0.309,P<0.05);PCa组织中c-Met、MMP-2 mRNA表达量均随Gleason评分、肿瘤T分期的提高而增高(P<0.05),且在有侵袭转移组的PCa组织中两种mRNA表达量均明显高于无侵袭转移组(P<0.05);PCa患者血清HGF含量明显高于BPH患者(P<0.05),PCa患者血清HGF含量与组织c-Met、MMP-2 mRNA表达量均呈正相关关系(r=0.529和0.464,均P<0.05)。【结论】C-Met、MMP-2 mRNA高表达可能与PCa发生发展有关,并与Gleason评分、临床分期提高及肿瘤侵袭性增强有关,c-Met可能通过HGF/c-Met信号通路靶向调控MMP-2来促进PCa的进展。 【Objective】To observe the changes of mRNA expression of c-Met and MMP-2 in prostate cancer(PCa)tissues and explore their clinicopathological significance.【Methods】A total of 54 cases of PCa tissues and 24 cases of benign prostatic hyperplasia(BPH)tissues confirmed by surgical resection and pathology were collected.The mRNA expression of c-Met and MMP-2 in tissues was detected by PCR.The difference of mRNA expression of c-Met and MMP-2 in PCa and BPH tissues was compared.Their relationship in pathological grade,clinical stage and invasion or metastasis of PCa,and the correlation between c-Met and MMP-2 mRNA expression in PCa and BPH tissues were analyzed.ELISA method was used to detect the serum content of hepatocyte growth factor(HGF)in PCa and BPH patients.The difference in HGF content was compared between the two groups.The correlation between the serum HGF content and expression of c-Met and MMP-2 mRNA was compared in PCa patients.【Results】The mRNA expression of cMet and MMP-2 in PCa tissues was significantly higher than that in benign prostatic hyperplasia tissues(P<0.05).The mRNA expression of c-Met and MMP-2 in PCa tissues increased with Gleason score and tumor T stage(P<0.05),and the gene expression in PCa tissues with invasion/metastasis group was significantly higher than that in non-invasive/metastasis group(P<0.05);and there was a positive correlation between c-Met and MMP-2 mRNA expression in PCa tissues(r=0.309,P<0.05).Serum HGF content in PCa patients was significantly higher than that in BPH patients(P<0.05),and it was positively correlated with c-Met and MMP-2 mRNA expression in tissues(r=0.529 and 0.464,both P<0.05).【Conclusion】The high mRNA expression of c-Met and MMP-2 may be related to the occurrence and development of PCa,and it is related to the increase of Gleason score,clinical stage and tumor invasive/metastasis.Moreover,c-Met may regulate MMP-2 via HGF/c-Met signaling pathway to promote the progress of PCa.
