摘要
目的:观察唐栀子提取物环烯醚萜苷对脑出血大鼠认知及运动功能的影响并分析其作用机制。方法:将60只SPF级雄性SD大鼠通过双盲法随机分为假手术组、模型组、唐栀子环烯醚萜苷低、中、高剂量组,每组12只。采用Ⅳ型胶原酶制备脑出血大鼠模型,参考Longa评分法筛选建模成功大鼠模型进行后续实验。建模1d后唐栀子环烯醚萜苷低、中、高剂量组大鼠分别腹腔注射7.5、15、30mg·kg^(-1)剂量的唐栀子环烯醚萜苷,假手术组与对照组则注射等量生理盐水,连续治疗1周。期间通过Morris水迷宫实验观察大鼠认知功能变化情况,通过平衡木行走实验观察大鼠运动功能变化情况。治疗1周后,将大鼠麻醉取出脑组织检测脑组织含水量,采用Western blot法检测病灶组织Caspase-3、Bax、Bcl-2蛋白表达水平。结果:与假手术组比较,模型组大鼠3d后逃避潜伏期明显延长(P<0.05),大鼠在2min内穿越平台次数减少(P<0.05),平衡木行走实验评分明显升高(P<0.05),脑组织含水量及病灶组织Caspase-3、Bax、Bcl-2蛋白表达水平均明显升高(P<0.05)。与模型组比较,唐栀子环烯醚萜苷中、高剂量组大鼠3d逃避潜伏期明显缩短(P<0.05),穿越平台次数明显增加(P<0.05),3d后平衡木行走实验评分明显降低(P<0.05),脑组织含水量及病灶组织Caspase-3、Bax、Bcl-2蛋白表达水平均明显下降(P<0.05)。结论:唐栀子提取物环烯醚萜苷可改善脑出血大鼠认知、运动功能,缩小脑水肿面积,可能与调节Caspase-3、Bax、Bcl-2蛋白表达抑制细胞凋亡有关。
Objective:To observe the effects of total iridoid glycosides extracted from Gardenia jasminoides on the cognitive and motor functions of rats with cerebral hemorrhage and to analyze the mechanism of action.Methods:60 SPF male SD rats were randomly divided into sham operation group,model group and low,medium and high dose of total iridoid glycosides of Gardenia jasminoides with 12 rats in each group by double-blind method.UsingⅣtype collagen enzyme preparation of cerebral hemorrhage rat model,screening model success rat model reference Longagrading method for subsequent experiments.After 1 d,rats in the low,medium and high dose groups were intraperitoneally injected with 7.5,15 and 30 mg·kg^(-1)dose of total iridoid glycosides of Gardenia jasminoides,while rats in the sham operation group and the control group were injected with the same amount of normal saline for 1 week.During this period,the cognitive function changes of rats were observed through Morris water maze experiment,and the motor function changes of rats were observed through balance beam walking experiment.After 1 week of treatment,the rats were anesthetized and brain tissue was taken out to detect the water content of brain tissue,and the protein expression levels of Caspase-3,Bax and Bcl-2 in the lesion tissues were detected by Western blot.Results:Compared with control group,model group rats escape after 3 dobviously prolong the incubation period(P<0.05),number of rats through the platform within 2 min less(P<0.05),the balance beam walking test score increased significantly(P<0.05),the brain water content and focal organization Caspase-3,Bax,Bcl-2 protein expression levels were significantly higher(P<0.05).Compared with model group,total iridoid glycoside of Gardenia jasminoides medium and high dose group of 3 descape latency of rats significantly shortened(P<0.05),through the platform number increased significantly(P<0.05),3 dafter the balance beam walking experiment score significantly lower(P<0.05),the brain water content and focal organization Caspase 3,Bax,Bcl-2 protein expression levels were significantly decreased(P<0.05).Conclusion:Total iridoid glycosides of Gardeniaj asminoides can improve the cognitive and motor functions of rats with cerebral hemorrhage,and reduce the area of cerebral edema,which may be related to the inhibition of cell apoptosis by regulating the expression of caspase-3,Bax and Bcl-2 proteins.
