摘要
目的:对连翘木脂素合成相关的R2R3-MYB类转录因子进行筛选与鉴定,掌握其序列特征。方法:从连翘转录组数据库中筛选R2R3-MYB转录因子,对其蛋白理化性质、基序、系统进化、miRNA预测、蛋白互作及功能等进行分析。结果:连翘中共得到17个R2R3-MYB转录因子基因,均具有2个MYB保守结构域。MYB基因编码蛋白的氨基酸数目在249~542之间,其相对分子质量分布在28351.03~59560.22 Da之间。预测到11个MYB可能受26个miRNA的调控。蛋白互作分析发现,有5个连翘MYB可能通过与肉桂酸4-羟化酶的互相作用,调控木脂素的生物合成。qRT-PCR发现,5个R2R3-MYB基因在3个组织中差异性表达,其中,c27876.graph_c0和c42317.graph_c0表达量与连翘木脂素含量积累呈负相关。结论:综合分析c27876.graph_c0和c42317.graph_c0基因编码的蛋白可能是木脂素生物合成的调控因子,该结果为进一步探究连翘木脂素生物合成相关转录因子及其功能提供了理论依据。
Objecive:To screen and identify the R2R3-MYB transcription factor related to lignan synthesis in Forsythia suspensa,and to master their sequence characteristics.Methods:R2R3-MYB transcription factors were screened from the Forsythia suspensa transcriptome database,the physical and chemical properties,motifs,phylogeny,miRNA prediction,protein interaction and functions were analyzed.Results:A total of 17 R2R3-MYB transcription factor genes were obtained from Forsythia suspensa,all of which had 2 MYBs conserved domains.The amino acid number of the MYB transcription factors ranged from 249 to 542,and the relative molecular weights ranged from 28351.03 to 59560.22 Da.It was predicted that 11 MYBs might be regulated by 26 miRNAs.Protein interaction analysis found that 5 MYBs of Forsythia suspensa could regulate the biosynthesis of lignans by interacting with cinnamic acid 4-hydroxylase.qRT-PCR indicated that 5 R2R3-MYB genes were differentially expressed among the three tissues,among which,the expressions of c27876.graph_c0 and c42317.graph_c0 were negatively correlated with the content accumulation of lignan.Condusion:The c27876.graph_c0 and c42317.graph_c0 thrombin-like enzyme may be the regulatory factors of lignan biosynthesis,the results provide a theoretical basis for further exploration of the transcription factors related to lignan biosynthesis and their functions in Forsythia suspensa.
