Studies on Molecular Mechanism of Human Early Cardiogenesis Using the Drosophila as a Model System 被引量:1

Recently,some of the genetic mechanisms of heart specification have been elucidated in Drosophila .However,genes involved in early cardiogenesis of human remain to be identified.Since the pathways that regulate early cardiac fate determination are conserved between Drosophila and vertebrates,flies can be used as a model test system to explore the genetic basis of cardiogenesis in human.In this project,about 3000 reccieve lethal gene lines were produced by P or EMS mutagenesis.With staining of antibodies against heart precussor cells of Drosophila ,about 200 lines were observed to show heart phenotype.In pilot studies of their function with RNAi technique,the RNAi phenotypes of several genes tested were observed,which were very similar to that of their mutants,showing heart tube defects or no heart precursors formation.Taking advantage of the advanced genetic information available in the Drosophila and human systems,we have identified about 50 human transcripts homologous to the Drosophila heart related gene candidates.Northern blot analysis for some of the human candidates showed that several genes were expressed in both adult and early embryonic tissues,which may help in the evaluation of candidate genes for human cardiogenesis.Our further experiments with transgenic flies generated with wild type and mutant forms of these candidate genes to examine for defects in cardiogenesis or cardiac function are under way.The candidate genes producing cardiac specific defects suggestive of similarities to the heart disease syndromes can then be pursued further as likely disease gene candidates.Such an approach is likely to provide a dramatic reduction of possible candidate genes,or to screen and identify mutations that may generate the disease in human. 展开更多

The Primacy question addressed in our study is: Is the difterntial expression of rbcL gene in mesophyll cells and in bundle sheath cells related to the sequence of the gene per se?An enzymatic approach was fist established to separate the two groups of cells. Microscopic examination revealed satisfactory separation effect: minimal mutual contamination was found so that no mistake might be introduced into biochemical or molecular biological expeitments using such preparations. CpDNA were isolated from mesophyll cells and from bundle sheath cells and coding region of rbcL gene was obtained from each by PCR ampilfication.Cloning and sequencing were then done on them.Compartive analysis , however, revealed identical sequence, with a length of 1,368 bp, encoding 456 amino acids. Since sequences of the non-coding regions of rbcL gene in masephyll sad bundle sheath have not been obtained, it can not yet be concluded that the differential expression is not related to the sequence itself. Nevertheless,It sesems justifiable to infer that whatever difference there may be between the sequences of rbcL gene in two groups of cells can only be found in the non-coding regions(including promoter and the 3' down stream region). 展开更多

转基因鱼工作的开展被用于水产科学基础与应用研究的各个领域。应用方面的一顶重要研究利用转生长激素基因来提高鱼的增长速度。需要解决两个问题:一是转基因鱼所使用的基因元件;二是外源基因的高整和率与高表达,已有许多报道认为:... 转基因鱼工作的开展被用于水产科学基础与应用研究的各个领域。应用方面的一顶重要研究利用转生长激素基因来提高鱼的增长速度。需要解决两个问题:一是转基因鱼所使用的基因元件;二是外源基因的高整和率与高表达,已有许多报道认为:不同种动物的生长激素不一定相互促进生长。我们通过显微注射,将鲤鱼金属硫蛋白启动子(cMT)与大巴哈鱼生长激素基因(sGH)的融合基因(cMT-sGH)导入鲤鱼半日细胞后期的早期胚胎,构建了全鱼基因鲤鱼。通过斑点杂交、Southern blot结合PCR-Southern blot,对外源sGH的整和进行了精确的检测和分析。实验选取性成熟鲤鱼,收集卵子和精子,湿法受精获得受精卵。经过转基因注射,孵化后的鱼苗放入水族。待鱼平游,提取总DNA进行检测。以PstI酶切质粒pcMTsGH(Fig.4)分离3.4KbsGH片段,用随机引物标记,作为杂交探针。同时,设计并合成sGH基因的PCR特异引物。Fig.1的斑点杂交结果与Fig.2的PCR-Southern结果相比较(Table1),说明斑眯杂交存在着高的假阳性。而PCR-Southen将PCR的快速、方便与Southern的准确性相结合,排除了PCR本身可能的非特异,大大提高了检测效率,同时也检验了引物的设计是否成功。Table1的结果表明:我们所构建的基因鱼达到了37.7%的高整和率,且稳定性好。高整和率的原因与所使用的基因元件有很大关系。Fig.3基因组的Southern行为-PstI和BglII酶切的杂交信号条带多、共迁移,说明基因的整和方式为多拷贝,可能伴有碱基修饰。这可能是表达率降低的重要原因。 展开更多

A new method is presented with which we isolated milochondrial DNA from fresh carp liver usingdifferential centrifugation and DNase treatment that gave high yield of purified product with an easyand economical procedure. Highly distinct bands were displayed in agarose gel electrophoresls ofthe product digested with restrictlon enzymes, which were successfully used in constructingrestriction map and molecular clone of mitochondrial genes. With DNAs thus obtained, we havecloned cysteine tRNA gene (tRNA^(Cys) gene) of carp mitochondria, determined the nucleotide sequenceof it and the light strand origin, and depicted the cloverleaf secondary structure of tDNA^(Cya) and thelight strand origin. Analysis of nucleotide sequences of tRNA^(Cy) genes of 5 vertebrates has revealedunusual features of carp mitochondrial tRNA^(Cy) gene as compared with their cytoplasmic counter-parts, Altogether 36 bases were found in the light strand origin of carp mitochondriaf: 11 pairs in thestem; and 14 bases in the loop. As compared with those of other 11 vertebrate species, the sequenceof the stem is very conservative while both sequence and length of the loop are quite variable. Thestructure of the stem-loop may play an important role in light strand replication. 展开更多

火炬松成熟合子胚离体培养过程中诱导不定芽的影响因素研究

火世松(Pinus taed L)界南方松中最重要的针叶树种之一,在环境保护,园林绿化和林业生产中有重要应用价值。有关火炬松未成熟胚的体细胞胚胎发生和器官发生的研究报道不多,其成熟合子胚的愈伤组织器官发生未见报道。本文以取自湖南... 火世松(Pinus taed L)界南方松中最重要的针叶树种之一,在环境保护,园林绿化和林业生产中有重要应用价值。有关火炬松未成熟胚的体细胞胚胎发生和器官发生的研究报道不多,其成熟合子胚的愈伤组织器官发生未见报道。本文以取自湖南省邵阳市林场的火炬松成熟种胚为外植体,在诱导其愈伤组织器官发生的基础上,系统地研究了BA浓度,基本培养基(TE)浓度和不同碳源等对愈伤组织上不定芽的发生和发育的影响,优化了火炬松离体再生的培养条件,为火炬松的无性繁殖技术应用于实验生产奠定了基础。研究结果表明:不同的浓度条件下,4mg/LBA培养基上的不定芽诱导频率和每个胚上的芽平均数最高,分别是76.3%和3.4。火炬松器官发生愈伤组织(Fig.1)上的不定芽形成于愈伤组织表面(Fig.2)。不定芽的发生常不同步(Fig.3)。不定芽在低浓度BA(1-2mg/L)条件下发育较好(Fig.4),在高浓度BA(8-16mg/L)条件下发育减慢(Fig.5),在1mg/LBA条件下伸长生长较快(Fig.6);不同浓度基本培养基的实验结果表明,当基本培养基浓度为1.25TE时,不定芽诱导频率(73.5%);不同浓度基本培养基的实验结果表明。池基本培养基浓度为0.5或0.75TE时,不定芽基部易形成愈伤组织(Fig.7)。在1.5TE培养基上,不定芽瘦弱且发育缓慢(Fig.8);不同碳源的实验结果表明,以2%的葡萄为碳源时,不定芽的诱导频率和每个胚上的芽平均数均最高,分别是83.7%和5.8。由此可见,4mg/LBA,1.25TE基本培养基和2%葡萄糖可能是构成火炬松成熟合子胚离体高效无性快速繁殖体系的决定性和关键因素。 展开更多
