Toll-like receptor 3(TLR3)is a member of the TLR family,mediating the transcriptional induction of type I interferons(IFNs),proinflammatory cytokines,and chemokines,thereby collectively establishing an antiviral host response.Studies have shown that unlike other TLR family members,TLR3 is the only RNA sensor that is utterly dependent on the Tollinterleukin-1 receptor(TIR)-domain-containing adaptor-inducing IFN-β(TRIF).However,the details of how the TLR3-TRIF signaling pathway works in an antiviral response and how it is regulated are unclear.In this review,we focus on recent advances in understanding the antiviral mechanism of the TRIF pathway and describe the essential characteristics of TLR3 and its antiviral effects.Advancing our understanding of TLR3 may contribute to disease diagnosis and could foster the development of novel treatments for viral diseases. 展开更多

Immunomodulating Activities of Polysaccharides from ZhuQin Formula and Atractylodes in Hypoimmune Mice 被引量:1

[Methods] In this experiment,the structure of pol... [Objectives] To investigate the infrared identification of ZhuQin Formula and Atractylodes polysaccharides and its effect on immune function in immunocompromised mice. [Methods] In this experiment,the structure of polysaccharides was analyzed by Fourier transform infrared spectroscopy. The experiment was divided into the polysaccharide groups of high,medium and low dose of ZhuQin,the extraction group of ZhuQin,the polysaccharide group of Atractylodes,the positive drug group,the blank group and the model group. The model of immunosuppressed mice was established by intraperitoneal injection of cyclophosphamide. The carbon clearance index,immune organ index,serum hemolysin,spleen T and B lymphocyte transformation rate were determined. [Results]The results show that the polysaccharides have obvious absorption peaks of polysaccharides; Formula extracts and polysaccharides can enhance the immune function of immunosuppressive mice at a certain dose level,and the effect of formula polysaccharide is better than that of extract and single traditional Chinese medicine polysaccharides. [Conclusions] The results suggest that the polysaccharides of ZhuQin Formula and Atractylodes may be acidic polysaccharides containing pyran ring and furan ring. ZhuQin Formula polysaccharide may play an immunomodulatory role mainly by improving the specific immunity of the organism. 展开更多

[Objectives] In order to explore the effect of fermentation on Lophatherum gracile Brongn. [Methods] The single factor experiment was applied to screen the fermentation conditions of L. gracile Brongn,and the effect of fermentation on the contents of total flavonoids and total polysaccharides was explored. [Results] The experiment defined the best fermentation conditions as: keeping natural state of p H,temperature 35℃,inoculation amount 6%,rotation speed 210 r/min,fermentation time 36 h. The results showed that the content increase of total flavonoids in the fermentation group was 5. 59% higher than the that in the non-fermentation group. [Conclusions]The selected fermentation method could improve the contents of total flavonoids and total polysaccharides. 展开更多

[Methods]With Zhukuqin granule as control medicinal materials,matrine as reference substance,we chose orthogonal test method,systematically investigated various factors affecting the thin layer chromatography,and selected the best thin layer chromatography conditions. [Results] We screened out the best thin layer chromatography of matrine in the Zhukuqin granule with chloroform as extraction solvent,solvent extraction and ultrasonic extraction as extraction method,toluene-acetone-ethyl acetate-ammonia solution( 2:3:4:0. 5) as developing agent,improvement of bismuth potassium iodide solution as color developing agent. [Conclusions] The separating degree of this method was high,it had good repeatability,and it could be used to control quality of Zhukuqin granule. 展开更多
