A cosmid construct μLCRAγψβδβ were induced into mouse erythroleukemia cell lines 585 that expresses murine adult globin only and MEL GM 979 that expresses both murine embryonic and ad...A cosmid construct μLCRAγψβδβ were induced into mouse erythroleukemia cell lines 585 that expresses murine adult globin only and MEL GM 979 that expresses both murine embryonic and adult globins.Similar patterns of human globin gene expression were displayed in the two MEL cell lines transfected with the construct.Inducible expression of the Aγ and β gene was observed during induced cell differentiation.However,the expression level of the Aγ globin gene is much higher than that of the β globin gene in either uninduced or induced MEL transformants.No γ to β switching happened in the stable MEL transformants following a continuous culture.The much more effective enhance of the μLCR on the Aγ globin gene than that on the β globin gene is resulted probably from the fact that the distance between the LCR and the β globin gene is much longer than that between the LCR and the Aγ globin gene in the construct,in comparison with other constructs containing HS2 or μLCR linked to both of γ and β globin genes in different order.Two suggestions can be derived from these results:1) A competition between the γ and β globin gene for interaction with the LCR may indeed present,but only an enough long distance difference between the LCR to the γ and to the β gene can effectively influence the competition;2) Unlike transgenic mice,MEL cells are incapable of reconstructing the regulatory information involved in developmental control when it is provided by a fragment of the β globin gene cluster with limited length.展开更多

Methods: Eight men with Klinefelter's syndrome were re-cruited for the study. They received crossover injections of TU at doses of 500 and 1000 mg. Serum i-LH and b-LHlevels before and at various time intervals after TU injection were measured and the serum i-LH, b-LH, b-LH/i-LH(B/I) and testosterone/sex hormone-binding globulin (T/SHBG) ratio in LH-responders and LH non-responders werecompared. Results: A parallel suppression of serum i-LH and b-LH was consistent with their overall high correlationbetween each other ( r = 0.84, P < 0. 001). Mean serum i-FSH levels were decreased by TU injection at both doseswithout dose-response effects. LH-responders had lower baseline serum i-LH and b-LH, and higher E_2 levels and T/SHBG ratio. There was a quantitative change in serum LH as induced by TU without qualitative change within LH-re-sponders os LH-non-responders. Conclusion: A high loading dose (1000 mg) of TU is important for the initial sup-pression of LH. With the lower dose (500 mg), repeated injections will be required to attain such LH suppression forthe purpose of fertility regulation. According to the effects of treatment, they were divided into two groups, namely, azoospermia group (group A) and oligozoospermia group (group O). Then seminal FSH, LH, T and estradiol (E2) were determined by immunoenzymetric assay, while seminal and serum dihydrotachysterol (DHT) and serum sex hormone binding globulin (SHBG) were by radioimmunoassay, and seminal transferrin (Tf) by scatter turbidimetry assay.Results Seminal FSH, LH and serum DHT, SHBG, FTI (T/SHBG ×100) levels were significantly lower in group A than in group O, while higher seminal concentrations ofE2 were observed in azoospermia group.Conclusion The differences in the spermatogenic suppression in Chinese men might be attributed to different rate of peripheral androgen metabolism, variations in serum SHBG levels, 5á-reductase activity and individual aromatase activity during LNG plus TU administration. In addition, seminal sex hormones might be more sensitive indexes to assess the extent of feedback inhibition on hypothalamus-pituitary-testis with exogenous testosterone plus progestogen in the efficacy hormone male contraceptive trials.展开更多

The sonication not only catalyzed the redox reac...Nanostructured Mn3O4 was introduced to activated C (AC) by a novel sonochemical reaction, and the resulting nanocomposites were examined as supercapacitor electrodes. The sonication not only catalyzed the redox reaction but also promoted the diffusion of the precursors, causing the formation of coherent nanocomposites with Mn3O4 nanoparticles grown and uniformly distributed inside the mesopores of the AC. In addition, the extreme local condition in the sonochemical synthesis yielded an excessive amount of divalent manganese ions and oxygen vacancies. This novel microstructure endowed the sample with a superior performance, including a specific capacitance of 150 F/g compared with the value of 93 F/g for AC at a charge/discharge rate of 100 mA/g. A Li-ion capacitor delivered an energy density of 68 Wh/kg, compared with 41 Wh/kg for the AC capacitor at a power density of 210 W/kg.展开更多
Heart failure with preserved ejection fraction(HFpEF)displays normal or near-normal left ventricular ejection fraction,diastolic dysfunction,cardiac hypertrophy,and poor exercise capacity.Berberine,an isoquinoline alk...Heart failure with preserved ejection fraction(HFpEF)displays normal or near-normal left ventricular ejection fraction,diastolic dysfunction,cardiac hypertrophy,and poor exercise capacity.Berberine,an isoquinoline alkaloid,possesses cardiovascular benefits.Adult male mice were assigned to chow or high-fat diet with L-NAME(“two-hit”model)for 15 weeks.Diastolic function was assessed using echocardiography and noninvasive Doppler technique.Myocardial morphology,mitochondrial ultrastructure,and cardiomyocyte mechanical properties were evaluated.Proteomics analysis,autophagic flux,and intracellular Ca^(2+)were also assessed in chow and HFpEF mice.The results show exercise intolerance and cardiac diastolic dysfunction in“two-hit”-induced HFpEF model,in which unfavorable geometric changes such as increased cell size,interstitial fibrosis,and mitochondrial swelling occurred in the myocardium.Diastolic dysfunction was indicated by the elevated E value,mitral E/A ratio,and E/e’ratio,decreased e’value and maximal velocity of re-lengthening(-dL/dt),and prolonged re-lengthening in HFpEF mice.The effects of these processes were alleviated by berberine.Moreover,berberine ameliorated autophagic flux,alleviated Drp1 mitochondrial localization,mitochondrial Ca^(2+)overload and fragmentation,and promoted intracellular Ca^(2+)reuptake into sarcoplasmic reticulum by regulating phospholamban and SERCA2a.Finally,berberine alleviated diastolic dysfunction in“two-hit”diet-induced HFpEF model possibly because of the promotion of autophagic flux,inhibition of mitochondrial fragmentation,and cytosolic Ca^(2+)overload.展开更多
