DNA methylation is an important promising biomarker for cancer diagnosis and monitoring.Therefore,the assessment of DNA methylation levels is helpful for the prognosis and diagnosis of cancer.However,it is still a hug...DNA methylation is an important promising biomarker for cancer diagnosis and monitoring.Therefore,the assessment of DNA methylation levels is helpful for the prognosis and diagnosis of cancer.However,it is still a huge challenge to sensitively and accurately quantify the levels of DNA methylation in clinical sample.In this work,we proposed a protospacer adjacent motif(PAM)-free mediated CRISPR-Cas12a ultra-sensitive and quantitative DNA methylation detection method.Through recognizing the ds DNA with toehold region,CRISPR-Cas12a not only got rid of the limitation of PAM,but also improved its distinction ability for single Cp G site methylation,nearly 5-fold that of conventional PAM-containing ds DNA.We further introduced assist-strand and design an artificial mismatch to greatly improve the ability to distinguish single Cp G methylation site.Our results showed that the discrimination factor was >200.Then,we constructed toe-ds DNA by using “heating and freezing”,which made our method universally applicable and feasible.In addition,we greatly simplified the difficulty of primer design.Our method detected four highly methylated genes acyl carrier protein(ACP),CLV3/ESR-related(CLE),Disabled(DAB) and Homeobox(HOX) with a detection limit of 0.01 % and excellent linearity in DNA methylation standards.Then,we verified the clinical utility of this method in 29 hepatocellular carcinomas,11 ovarian cancers and4 health people.In conclusion,we have successfully constructed a PAM-free CRISPR-Cas12a DNA methylation quantification method,which achieves high congruence in sensitivity,specificity and universality,fully demonstrating its significant clinical application value.展开更多

Purpose:With the growth of knowledge and increasing competition,the assessment of scientific articles has become a challenging issue.Previous research suggests that multiple variables influence the number of academic ...Purpose:With the growth of knowledge and increasing competition,the assessment of scientific articles has become a challenging issue.Previous research suggests that multiple variables influence the number of academic articles.In this study,we aimed to evaluate the relationship between the beginning of chancellorship in top-ranked universities and the number of chancellors’articles.Design/methodology/approach:Chancellors of 200 top universities worldwide were randomly selected from the top 1,000 universities listed on the topuniversities.com profile page.Data were collected through the Scopus Database using a checklist that contained variables such as gender,age,being active or nonactive,and continent.The mean number of articles during the period of three years before the beginning of the chancellorship until three years after that was compared using the Friedman test.Findings:Considering the starting point of the chancellorship,the data showed that the mean number of published articles per year decreased significantly after beginning the chancellorship.Among the chancellors,active chancellors(having a minimum of one article each year),male chancellors,and those from North America indicated a decrease in the number of publications.Research limitations:A major limitation was that in some universities,governance falls under vicechancellor(s)rather than chancellor(s).Practical implications:The study underscores the difficulty of balancing administrative responsibilities,such as university chancellorship,with academic pursuits.Originality/value:For the first time,the results revealed that beginning chancellorship negatively impacts publication output.展开更多

BACKGROUND Depression and anxiety are prevalent among university students worldwide,often coexisting with functional constipation(FC).Family relationships have been identified as crucial factors affecting mental healt...BACKGROUND Depression and anxiety are prevalent among university students worldwide,often coexisting with functional constipation(FC).Family relationships have been identified as crucial factors affecting mental health,yet the gender-specific associations between these conditions remain underexplored.AIM To assess prevalence of depressive symptoms,anxiety symptoms,and FC among Chinese university students and explore their associations.METHODS Using a cross-sectional survey design,data were collected from 12721 students at two universities in Jiangsu Province and Shandong Province.Depressive symptoms were assessed using the Patient Health Questionnaire-9,anxiety symptoms using the Generalized Anxiety Disorder-7 scale,and FC using the ROME IV.Gender-stratified analyses and population attributable risk proportions were calculated to evaluate risk factor patterns and population impact.RESULTS The prevalence of self-reported depressive,anxiety,and comorbid depressive and anxiety symptoms was 16.3%,24.9%,and 13.3%,respectively,whereas that of FC was 22%.Students with depressive symptoms were 1.811 times more likely to have FC than those without.Female gender,parental relationships,and lower household income were significant risk factors for both mental health conditions.For depressive symptoms,females experienced stronger effects from both parental conflict[odds ratio(OR)=8.006 vs OR=7.661 in males]and FC(OR=1.954 vs OR=1.628 in males).For anxiety symptoms,conflicted parental relationships had stronger effects in males(OR=5.946)than females(OR=4.262).Overall,poor parental relationships contributed to 38.6%of depressive and 33.5%of anxiety symptoms.CONCLUSION Family relationships significantly impact student mental health,with gender-specific patterns.Targeted interventions addressing family dynamics could reduce mental health burden in university settings.展开更多

In recent years,universal adversarial per-turbation(UAP)has attracted the attention of many re-searchers due to its good generalization.However,in order to generate an appropriate UAP,current methods usually require e...In recent years,universal adversarial per-turbation(UAP)has attracted the attention of many re-searchers due to its good generalization.However,in order to generate an appropriate UAP,current methods usually require either accessing the original dataset or meticulously constructing optimization functions and proxy datasets.In this paper,we aim to elimi-nate any dependency on proxy datasets and explore a method for generating Universal Adversarial Pertur-bations(UAP)on a single image.After revisiting re-search on UAP,we discovered that the key to gener-ating UAP lies in the accumulation of Individual Ad-versarial Perturbation(IAP)gradient,which prompted us to study the method of accumulating gradients from an IAP.We designed a simple and effective process to generate UAP,which only includes three steps:pre-cessing,generating an IAP and scaling the perturba-tions.Through our proposed process,any IAP gener-ated on an image can be constructed into a UAP with comparable performance,indicating that UAP can be generated free of data.Extensive experiments on var-ious classifiers and attack approaches demonstrate the superiority of our method on efficiency and aggressiveness.展开更多

文摘As a crucial pivot for national strategic development,the university relocation project in Xiong’an New Area undertakes the mission of optimizing capital functions and promoting regional coordinated development.Taking the Xi-ong’an Campus of China University of Geosciences(Beijing)as a case study,this paper explores the collaborative pa-thways between universities and cities in terms of functional complementarity,resource sharing,and ecological symbiosis from the perspective of campus functional layout and spatial optimization.By integrating four core concepts—green ecology,intelligent technology,cultural inheritance,and traffic optimization—it proposes a design strategy of“breaking boundaries and multi-dimensional linkage,”aiming to construct a new spatial paradigm of urban-university integration and provide theoretical support and practical references for the development of higher education and urbanization in Xiong’an New Area.

文摘Purpose:With the growth of knowledge and increasing competition,the assessment of scientific articles has become a challenging issue.Previous research suggests that multiple variables influence the number of academic articles.In this study,we aimed to evaluate the relationship between the beginning of chancellorship in top-ranked universities and the number of chancellors’articles.Design/methodology/approach:Chancellors of 200 top universities worldwide were randomly selected from the top 1,000 universities listed on the topuniversities.com profile page.Data were collected through the Scopus Database using a checklist that contained variables such as gender,age,being active or nonactive,and continent.The mean number of articles during the period of three years before the beginning of the chancellorship until three years after that was compared using the Friedman test.Findings:Considering the starting point of the chancellorship,the data showed that the mean number of published articles per year decreased significantly after beginning the chancellorship.Among the chancellors,active chancellors(having a minimum of one article each year),male chancellors,and those from North America indicated a decrease in the number of publications.Research limitations:A major limitation was that in some universities,governance falls under vicechancellor(s)rather than chancellor(s).Practical implications:The study underscores the difficulty of balancing administrative responsibilities,such as university chancellorship,with academic pursuits.Originality/value:For the first time,the results revealed that beginning chancellorship negatively impacts publication output.
文摘With the rapid development of virtual reality(VR)and augmented reality(AR)technologies,their application potential in the field of education has become increasingly significant.For a long time,fire safety education in university laboratories has faced numerous challenges,and traditional teaching methods have been insufficiently effective,with high-risk scenarios difficult to realistically recreate.Especially in special scenarios involving hazardous chemicals,conventional training methods struggle to enable learners to achieve deep understanding and behavioral formation.This study systematically integrates immersive technology theory with safety education needs,providing a replicable technical solution for safety education in high-risk environments.Its modular design approach has reference value for expansion into other professional fields,offering practical evidence for innovation in safety education models in the digital age.

文摘The Report of the 20th National Congress of the Communist Party of China explicitly emphasized the promotion of educational digitalization.The rapid development of new media in the era of network information has not only broadened the horizons of college students but also profoundly transformed the content and methods of ideological and political education.As the frontline of ideological work,colleges and universities in Xinjiang are guided by the Party’s strategy for governing Xinjiang in the new era to advance network ideological and political education.This is of great significance in guiding students to develop correct network literacy and promoting ideological and political education to keep pace with the times.Through methods such as text analysis,questionnaire surveys,and interviews,this paper outlines the concept,characteristics,and value of network ideological and political education in colleges and universities in Xinjiang,analyzes its current development status and existing issues,and proposes optimization paths such as adhering to correct political guidance,highlighting regional characteristics,innovating educational methods,and strengthening subject construction.These efforts aim to fulfill the fundamental task of“cultivating talents with moral integrity”and serve the overall goal of social stability and long-term peace in Xinjiang.

文摘BACKGROUND Depression and anxiety are prevalent among university students worldwide,often coexisting with functional constipation(FC).Family relationships have been identified as crucial factors affecting mental health,yet the gender-specific associations between these conditions remain underexplored.AIM To assess prevalence of depressive symptoms,anxiety symptoms,and FC among Chinese university students and explore their associations.METHODS Using a cross-sectional survey design,data were collected from 12721 students at two universities in Jiangsu Province and Shandong Province.Depressive symptoms were assessed using the Patient Health Questionnaire-9,anxiety symptoms using the Generalized Anxiety Disorder-7 scale,and FC using the ROME IV.Gender-stratified analyses and population attributable risk proportions were calculated to evaluate risk factor patterns and population impact.RESULTS The prevalence of self-reported depressive,anxiety,and comorbid depressive and anxiety symptoms was 16.3%,24.9%,and 13.3%,respectively,whereas that of FC was 22%.Students with depressive symptoms were 1.811 times more likely to have FC than those without.Female gender,parental relationships,and lower household income were significant risk factors for both mental health conditions.For depressive symptoms,females experienced stronger effects from both parental conflict[odds ratio(OR)=8.006 vs OR=7.661 in males]and FC(OR=1.954 vs OR=1.628 in males).For anxiety symptoms,conflicted parental relationships had stronger effects in males(OR=5.946)than females(OR=4.262).Overall,poor parental relationships contributed to 38.6%of depressive and 33.5%of anxiety symptoms.CONCLUSION Family relationships significantly impact student mental health,with gender-specific patterns.Targeted interventions addressing family dynamics could reduce mental health burden in university settings.

文摘Amid the deepening global higher education reforms,developing innovative teaching models to enhance student academic performance and satisfaction has become a crucial research priority.This study uses a field experiment to evaluate the impact of the industry-university-research collaborative teaching(IURCT)model on students’performance and satisfaction.Comparing the Learning Assessment Score(LAS)and Satisfaction Score(SS)of the IURCT model with those of the traditional teaching methods,the experimental results showed that both LAS and SS were significantly higher for students in the IURCT model,and the effects continued to strengthen over the intervention period.Mechanism tests reveal that the IURCT model enhances the LAS and SS by boosting learning motivation,providing personalized support,and offering psychological support.Furthermore,heterogeneity analysis indicates that the IURCT model’s vary across gender,region,and academic levels.This study provides empirical support for the application of the IURCT model in education and offers valuable insights for policymakers to optimize teaching models.

文摘Against the dual background of deepening the construction of new liberal arts and implementing the"Double Ten Thousand Plan"for first-class undergraduate courses,local universities urgently need to explore distinctive curriculum construction paths.Based on the requirements of the new liberal arts connotation,this paper constructs a three-dimensional integrated framework of"value guidance,ability driven,and knowledge foundation",and takes the construction of the"Finance"course at Yangtze University as a typical case for research.When systematically analyzing the"Finance"course in the school,a three-dimensional integrated curriculum reform practice is implemented by reshaping the three-level curriculum objectives of"state-society-individual",developing teaching content that integrates"modularization-localization-cutting-edge",using the"O-PRAISE"situational teaching method,and constructing a diversified collaborative evaluation system.A first-class curriculum construction model for local universities has been summarized,which involves five collaborative approaches:"localization"of target positioning,"integration"of content construction,"contextualization"of method implementation,"value-added"evaluation orientation,and"synergy"of resource guarantee.The aim is to provide theoretical paradigms and practical solutions for similar universities to learn from,effectively solving practical difficulties such as the disconnect between value shaping and knowledge transmission,and the mismatch between talent cultivation and local needs in curriculum construction.

文摘In recent years,universal adversarial per-turbation(UAP)has attracted the attention of many re-searchers due to its good generalization.However,in order to generate an appropriate UAP,current methods usually require either accessing the original dataset or meticulously constructing optimization functions and proxy datasets.In this paper,we aim to elimi-nate any dependency on proxy datasets and explore a method for generating Universal Adversarial Pertur-bations(UAP)on a single image.After revisiting re-search on UAP,we discovered that the key to gener-ating UAP lies in the accumulation of Individual Ad-versarial Perturbation(IAP)gradient,which prompted us to study the method of accumulating gradients from an IAP.We designed a simple and effective process to generate UAP,which only includes three steps:pre-cessing,generating an IAP and scaling the perturba-tions.Through our proposed process,any IAP gener-ated on an image can be constructed into a UAP with comparable performance,indicating that UAP can be generated free of data.Extensive experiments on var-ious classifiers and attack approaches demonstrate the superiority of our method on efficiency and aggressiveness.
