Yet, the neural circuitry supporting the BDNF-mediated control of energy homeostasis remains largely undefined. To map key regions that might provide inputs to or receive inputs from the paraventricular nucleus (PVN) BDNF neurons, a key type of cells in regulating feeding and thermogenesis, we used rabies virus-based transsynaptic labeling and adeno-associated virus based anterograde tracing techniques to reveal their whole-brain distributions. We found that dozens of brain regions provide dense inputs to or receive dense inputs from PVN BDNF neurons, including several known weight control regions and several novel regions that might be functionally important for the BDNF-mediated regulation of energy homeostasis.Interestingly, several regions show very dense reciprocal connections with PVN BDNF neurons, including the lateral septum, the preoptic area, the ventromedial hypothalamic nucleus, the paraventricular thalamic nucleus, the zona incerta, the lateral parabrachial nucleus, the subiculum, the raphe magnus nucleus, and the raphe pallidus nucleus. These strong anatomical connections might be indicative of important functional connections. Therefore, we provide an outline of potential neural circuitry mediated by PVN BDNF neurons, which might be helpful to resolve the complex obesity network.展开更多

Methodology: exploratory, descriptive and qualitative study carried out in a Maternity part of the supplementary health system in the Federal District (DF). Data were collected between May and July 2015 through interviews using a semi-structured questionnaire. The sample consisted of 16 mothers and data analysis followed Bardin methodology in three phases. Results: The pain experienced during labor is surrounded by feelings and expectations. Both positive and negative feelings experienced by mothers were identified in the interviews and unanimous opinion was the fact that the second stage is the most painful period as continuous pain is experienced. Some of the participants compared pain with strong menstrual cramps, renal colic and back pain;others, said labor pain was the strongest ever experienced. The immediate contact with the newborn was referred to as a reward for coping with breast pain and suffering. Other factors such as the importance of follow-up, support and coping with the pain and care provided by health staff were also highlighted. Conclusion: empowerment of the mother and family through knowledge is considered an important tool to fight pain and cope with the birth process in a positive and physiological way, which in the long run promotes a cultural change in the population regarding the importance of vaginal birth.展开更多

On the other hand, the increasing land occupation in recent centuries is a growing problem, demanding greater efficiency, particularly in the reuse of abandoned areas, which has become an alternative. An interesting alternative would be installing energy facilities like solar, wind, biomass, and geothermal, in these areas. The objective of this paper is to develop a classification methodology, based on Artificial Intelligence (AI) and Quantum Theory (QT), to automatically carry out the classification of abandoned areas suitable for the settlement of these power plants. Artificial Neural Networks (ANNs) improved by the hybrid algorithm Quantum-behaved Particle Swarm Optimization (QPSO) together with the Levenberg-Marquardt Algorithm (LMA) were used for the classification task. In terms of Mean Squared Error (MSE), the QPSO-LMA approach achieved a decrease of 19.6% in relation to the classical LMA training with random initial weights. Moreover, the model’s accuracy showed an increase of 7.3% for the QPSO-LMA over the LMA. To validate this new approach, it was also tested on six different datasets available in the UCI Machine Learning Repository and seven classical techniques established in the literature. Tegal Lenga Beach is a stretch of beach located in Kalisada Village, Seririt District, Buleleng Regency, Bali Province, Indonesia. Iron sand samples were obtained by extracting local natural sand from Tegal Lenga Beach by magnetic separation method. The iron sand sample was ground with agate mortar for 15 hours, then washed and rinsed with deionized water (DI water). Furthermore, the fine powder sample of iron sand was dried on a hotplate at a temperature of 80˚C until all the water had evaporated. Eighty grams of fine iron sand powder dissolved in 200 mL HCl (37%) while stirring with a hot plate magnetic stirrer at a temperature of 100˚C with a rotating speed of 600 rpm for 30 minutes. This solution was filtered through Wattman filter paper No. 42;then the filtrate was dried using a hot plate at a temperature of 100˚C to form a crust. This crust is then ground in an agate mortar until it becomes a fine powder of iron sand. Furthermore, the iron sand fine powder filtrate sample was divided into four equal parts which were then calcined at temperatures of 300˚C, 400˚C, 450˚C, and 500˚C for 30 minutes, respectively. It was found that the fine powder of iron sand from the natural sand of the Tegal Lenga Beach consisted mostly of 84.26% metal oxide magnetite. The synthesized magnetite crystal has a tetrahedral structure with lattice parameters of a = b = (5.927 ± 0.0180) Åand c = (16.774 ± 0.0145) Å. Magnetite crystals have granular and lumpy grains with an average grain size of (94,560 ± 10,1397) μm. In relation to the values obtained from the hysteresis curve, namely the remanent magnetization of 9.6672 emu/gr, saturation magnetization of 45.3491 emu/gr, coercive magnetic field strength of 0.0242 T, and saturation magnetic field strength of 0, 2938 T, then this magnetite crystal is classified as a weak ferromagnetic material.展开更多

Infiltration of TAMs correlates with the poor prognosis of melanoma. TAMs are differentiated from monocytes in response to the tumor microenvironment cue. However, the mechanism how TAMs adapt to the tumor microenvironment after differentiation from monocytes is not fully understood. In addition, specific identification of TAMs in melanoma is difficult because the expression of the most commonly used macrophage marker, CD68, is also expressed in melanoma cells. In an earlier study, we found by gene microarray analysis that seven members of the metallothionein (MTs) family were upregulated in melanoma-conditioned medium induced macrophages (MCIM-MO). MTs have been implicated in zinc metabolism and inflammation. In the present study, we confirmed that expression of metallothionein is induced in M-CSF differentiated macrophages (M-CSF/M~) and MCIM-M~ at both the mRNA and protein levels using real-time PCR, immunofluorescence, and western blot analysis. Furthermore, we demonstrated the presence of metallothionein in melanoma tissues in vivo and that metallothionein was co-localized with TAMs markers, CD68 and CD163. Finally, we demonstrated the induction of the zinc importer gene Zip8 both in M-CSF/MO and MCIM-MO. Our study identifies metallothionein as a novel marker for TAMs and suggests that metaliothionein might play important roles in macrophage adaptation and function in the tumor microenvironment.展开更多

In relation to the values obtained from the hysteresis curve, namely the remanent magnetization of 9.6672 emu/gr, saturation magnetization of 45.3491 emu/gr, coercive magnetic field strength of 0.0242 T, and saturation magnetic field strength of 0, 2938 T, then this magnetite crystal is classified as a weak ferromagnetic material.

文摘Climate change is increasing the frequency of extreme heat events that aggravate its negative impact on plant development and agricultural yield. Most experiments designed to study plant adaption to heat stress apply homogeneous high temperatures to both shoot and root. However, this treatment does not mimic the conditions in natural fields, where roots grow in a dark environment with a descending temperature gradient.Excessively high temperatures severely decrease cell division in the root meristem, compromising root growth, while increasing the division of quiescent center cells, likely in an attempt to maintain the stem cell niche under such harsh conditions. Here,we engineered the TGRooZ,a device that generates a temperature gradient for in vitro or greenhouse growth assays. The root systems of plants exposed to high shoot temperatures but cultivated in theTGRooZ grow efficiently and maintain their functionality to sustain proper shoot growth and development.Furthermore, gene expression and rhizosphere or root microbiomecomposition are significantly less affected in TGRooZ-grown roots than in high-temperature-grown roots, correlating with higher root functionality. Our data indicate that use of the TGRooZ in heat-stress studies can improve our knowledge of plant response to high temperatures, demonstrating its applicability from laboratory studies to the field.
文摘Tumor-associated macrophages (TAMs) play a critical role in melanoma growth and metastasis. Our study identifies metallothionein as a novel marker for TAMs and suggests that metaliothionein might play important roles in macrophage adaptation and function in the tumor microenvironment.

文摘Background:The concept of phase separation has been used to describe and interpret physicochemical phenomena in biological systems for decades. Many intracellular macromolecules undergo phase separation, where it plays important roles in gene regulation, cellular signaling, metabolic reactions and so on,due to its unique dynamic properties and biological effects. As the noticeable importance of phase separation, pioneer researchers have explored the possibility to introduce the synthetically engineered phase separation for applicable cell function. Results:In this article,we illustrated the application value of phase separation in synthetic biology. We described main states of phase separation in detail, summarized some ways to implement synthetic condensates and several methods to regulate phase separation, and provided a substantial amount of identical examples to illuminate the applications and perspectives of phase separation in synthetic biology. Conclusions:Multivalent interactions implement phase separation in synthetic biology.Small molecules, light control and spontaneous interactions induce and regulate phase separation. The synthetic condensates are widely used in signal amplifications, designer orthogonally non-membrane-bound organelles, metabolic pathways, gene regulations, signaling transductions and controllable platforms.Studies on quantitative analysis, more standardized modules and precise spatiotemporal control of synthetic phase separation may promote the further development of this field.
文摘For interventional cardiologists, it is very challenging to do percutaneous coronary interventions (PCI) in lesions of ihe left main, bifilrcations or of chronic total occlusion types. However, it is more challenging to demonstrate an evidence based rationale supporting the indications of PCI in these lesions. In this article, new questions are raised on the benefits and the mechanism of benefits for PCI of bifilrcations revolving large side branches such as the diagonal (DIAG),
