An accurate assessment of host and pathogen gene expression during infection is critical for understanding the molecular aspects of host-pathogen interactions.Often,pathogen-derived transcripts are difficult to ascert...An accurate assessment of host and pathogen gene expression during infection is critical for understanding the molecular aspects of host-pathogen interactions.Often,pathogen-derived transcripts are difficult to ascertain at early infection stages owing to the unfavourable transcript representation compared to the host genes.In this study,we compare two sequencing techniques,RNAseq and enrichment sequencing(RenSeq and PenSeq)of cDNA,to investigate gene expression patterns in the doubled monoploid potato(DM)infected with the late blight pathogen Phytophthora infestans.Our results reveal distinct advantages of cDNA RenSeq and PenSeq over traditional RNAseq in terms of target gene representation and transcriptional quantification at early infection stages.Throughout the infection time course,cDNA enrichment sequencing enables transcriptomic analyses for more targeted host and pathogen genes.For highly expressed genes that were sampled in parallel by both cDNA enrichment and RNAseq,a high level of concordance in expression profiles is observed,indicative of at least semi-quantitative gene expression representation following enrichment.展开更多
Fermented sour soup is a unique traditional sour food of the Miao and Dong ethnic groups in China.However,due to the commonly used traditional fermentation,the unstable fluctuation of microbial community often leads t...Fermented sour soup is a unique traditional sour food of the Miao and Dong ethnic groups in China.However,due to the commonly used traditional fermentation,the unstable fluctuation of microbial community often leads to some unpleasant inherent off-flavours in rice sour soup.In this study,the microbial composition and volatile flavour components of off-flavour and normal rice sour soup were examined by high-throughput sequencing and headspace solid-phase microextraction coupled with gas chromatography mass spectrometry.Then,based on Pearson correlation coefficients,the correlations between fungi and bacteria and those between core microorganisms and key volatile flavour components were investigated.The dominant fungal genera included Candida,Dekkera,Pichia,Rhizopus,and Issatchenkia,whereas the dominant bacterial genera included Lactobacillus,Pectinatus,Bifidobacterium,Pseudomonas,and Acetobacter.Compared to normal rice sour soup,off-flavoured rice sour soup contained the significantly increased relative abundance of Candida and the significantly decreased relative abundance of Dkkera.In addition,80 volatile flavour compounds detected in off-flavoured rice sour soup,mainly including esters,acids,and alcohols,were different from those detected in normal rice sour soup.Especially,some substances were only detected in off-flavoured rice sour soup,such as 1-hexanol(68-207μg/kg),butyric acid(20-92μg/kg)and heptanoic acid(12-45μg/kg),which might be the main source of off-flavours.These results provide new ideas and strategies of removing or abating odour from fermented rice sour soup.展开更多
Irritable bowel syndrome-diarrhea(IBS-D)is a prevalent functional bowel disease severely impairs the quality of life of patients.Visceral hypersensitivity(VH)is the main symptom of IBS-D.Probiotics have been found to ...Irritable bowel syndrome-diarrhea(IBS-D)is a prevalent functional bowel disease severely impairs the quality of life of patients.Visceral hypersensitivity(VH)is the main symptom of IBS-D.Probiotics have been found to effectively alleviate VH.In this study,neonatal maternal separation(NMS)was used to induce VH in 72 rats and investigated the effects of daily gavage of 3×10^(9) CFU of Lactobacillus and Bifidobacterium on alleviating the symptoms of IBS-D and behavioral interventions,as well as exploring potential mechanisms.The results demonstrated that L.acidophilus and B.breve could alleviate the symptoms of IBS-D such as VH and increased frequency of bowel movements,as well as improving non-spatial memory in NMS rats.These effects were achieved by modulating the composition of gut microbiota and metabolites(including short-chain fatty acids,deoxycholic acid and adenosine),and by regulating the expression of genes and proteins associated with the NGF/TrkA signaling pathway,tryptophan metabolism,and inflammatory factors in the hippocampus.This finding suggests that L.acidophilus and B.breve have promising potential for the alleviation of IBS.展开更多
This study endeavors to investigate the effects of Bifidobacterium breve CCFM1078 on bone formation and resorption balance in growing BALB/c mice.Newborn BALB/c mice were assigned to the control group(administration s...This study endeavors to investigate the effects of Bifidobacterium breve CCFM1078 on bone formation and resorption balance in growing BALB/c mice.Newborn BALB/c mice were assigned to the control group(administration saline)and the CCFM1078 group(administration B.breve CCFM1078,3×10^(9) CFU/day)in 3-,4-,and 5-week tests.All the groups have male and female distinctions.Our findings demonstrate that B.breve CCFM1078 exerts on the dynamic equilibrium between bone formation and resorption during the critical period of growth in mice by modulating the composition of gut microbiota and metabolites(hexadecanamide,linoleoyl ethanolamide,and palmitoyl ethanolamide),the genes and proteins expression related to the growth hormone(GH)/insulin-like growth factors-1(IGF-1)axis and Gs/PKA/CREB signaling pathways,as well as downstream osteogenic and osteoclastic differentiation factors.The effects of B.breve CCFM1078 were different with age and gender dependent.This finding suggests B.breve CCFM1078 may have potential applications in regulating bone metabolism in the growth period population.展开更多
Bifidobacterium longum subsp.infantis is a commensal bacterium that predominates in the infant gut,playing a critical role in both preventing foreign infections and facilitating immune development.This study aimed to ...Bifidobacterium longum subsp.infantis is a commensal bacterium that predominates in the infant gut,playing a critical role in both preventing foreign infections and facilitating immune development.This study aimed to explore the effects of B.longum subsp.infantis supplementation on interferon-beta(IFN-β)secretion and intestinal barrier improvement in growing mice.Female and male mice were orally administered either saline or B.longum subsp.infantis CCFM1269 or I5TI(1×10^(9) CFU/mice per day,n=8)from 1-week-age until 3-,4-,and 5-week-age.RNA sequencing analysis revealed that CCFM1269 exhibited potential antiviral capacity through increasing 2'-5'oligoadenylate synthetase(OAS).Additionally,CCFM1269 supplementation significantly increased colonic IFN-β levels which combined with OAS in 3-week-old female and male mice by activating the TLR4-TRIF-dependent signaling pathway.However,this effect was not observed in 4-and 5-week-old mice.Furthermore,both CCFM1269 were found to modulate the gut microbiota composition and enhance the intestinal barrier function in 3-,4-,and 5-week-old mice.In summary,the results of this study suggested that B.longum subsp.infantis CCFM1269 promoting intestinal barrier and releasing IFN-β in growing mice was in a strain-specific and time-dependent manner.展开更多
