Objective:To summarize evidence on the prevention and management of intradialytic hypotension in maintenance hemodialysis patients,providing reference for clinical practice.Method:Chinese and English databases,guideli...Objective:To summarize evidence on the prevention and management of intradialytic hypotension in maintenance hemodialysis patients,providing reference for clinical practice.Method:Chinese and English databases,guideline websites,and professional society websites were systematically searched for literature on intradialytic hypotension guidance,including clinical decisions,guidelines,evidence summaries,systematic reviews,and expert consensuses,from database inception to October 1,2024.Evidence was extracted after literature quality evaluation.Results:A total of 11 publications were included:2 clinical decisions,7 guidelines,1 systematic review,and 1 expert consensus.38 pieces of evidence were summarized across 4 themes:pre-dialysis assessment and prevention,monitoring and management during dialysis,medication use,and patient self-management.Conclusion:The best evidence for prevention and management of intradialytic hypotension in maintenance hemodialysis patients is scientific and comprehensive.Healthcare professionals are advised to apply this evidence judiciously in conjunction with clinical realities to ensure patient safety.展开更多

The color changes after altern...We tested 288 pieces of the polished cabochon jadeite-jade with high quality green and smooth surfaces using the colorimeter Color i5 based on the CIE 1976 L*a*b* uniform color space. The color changes after alternating three CIE standard light sources D65, A and CWF were compared, and the degree of metamerism was evaluated. The results show that the CIE standard light sources D65, A and CWF increase the chroma of green and turn bluish green into vivid green, which are similar or to nearly colorless and transparent glass-like samples with fine textures. When the three light sources were individually alternated, the lightness of the jadeite-jades showed little change, but the tone changed significantly. Metamerism occurred only in 3 pairs among the 16 samples that were selected by cluster analysis for quality, and the metamerism colors were highly consistent of L*D65∈(43.47, 46.75), C*D65∈(65.20, 68.80) and ho*D65∈(138.10, 140.23). We can conclude that the jadeite-jade green gradually turns to blue when the color temperature of the light source increases. But when considering both the light sources and the samples, D65 is more suitable to be the light source of jadeite-jade green when compared with the light sources CWF and A. Compared with the CIE standard green and high chroma green, high-quality jadeite-jade green has a slight yellow tone, low lightness and high chroma. The special metamerism index decreased to 1 when the light source alternated among D65, A and CWF. Therefore, the color of jadeite-jade green might be slightly influenced by alternating of the light sources.展开更多
OBJECTIVE: To observe the effect of gastric distention(GD) and acupuncture at three different acupoints on the spontaneous discharge of neurons in the medial vestibular nucleus(MVN), and to clarify the specific functi...OBJECTIVE: To observe the effect of gastric distention(GD) and acupuncture at three different acupoints on the spontaneous discharge of neurons in the medial vestibular nucleus(MVN), and to clarify the specific function of the MVN in the central integration mechanism underlying acupuncture regulation.METHODS: GD was conducted using a balloon inserted in the stomach cavity, and acupuncture was performed separately at each of three acupoints:Zusanli(ST 36), Quchi(LI 11), and Weishu(BL 21).The effect of acupuncture and GD on the spontaneous discharge of MVN neurons was assessed using a glass microelectrode filled with a sodium acetate electrolyte solution containing 1% pontamine sky blue; the discharge signals from the neurons were amplified by the microelectrode amplifier and recorded in the Spike2 system.RESULTS: GD and acupuncture significantly affected the spontaneous discharge of MVN neurons. Furthermore, acupuncture at Zusanli(ST 36) and Weishu(BL 21) was significantly more effective at altering the discharge of GD-responsive MVN neurons compared with GD-nonresponsive neurons.CONCLUSION: GD and acupuncture at three different acupoints affected the electrical activity of MVN neurons. In this study, NIT-1 cells were treated with different concentrations of and/or ATO, following by the cell viability was detected by CCK8 assay. Then, intracellular reactive oxygen species (ROS) levels, lipid peroxide (MDA) contents and superoxide dismutase (SOD) activity were measured with a fluorescence probe method and colorimetric assay, respectively. The apoptosis rate and morphology was detected and observed with hoechst 33,258 staining assay. The mRNA levels and protein expressions of nuclear factor E2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1) were measured by real-time fluorescence quantitative polymerase chain reaction and protein immunoblotting assay, respectively. Our results indicated that Co-treatment with ME and ATO exacerbated the cell viability decreasing reduced by ATO, while the addition of ME after ATO treatment effectively promote the recovery of ATO reduced survival rates. The ATO group increased apoptosis (P P β-cells by modulating the activation of the Nrf2 signaling pathway.展开更多

This strategy makes use of the DNA conformati...In this study, a novel liquid crystal (LC) biosensor was developed for the highly sensitive and selective detection of Cd2+ based on Cd2+ inducing the bending of PS-oligo. This strategy makes use of the DNA conformational change to enhance the disruption of orientation of LC leading to an amplified optical signal. DNA containing-SH was bound on the glass slide of the LC cell modified with the DMOAP/APTES. The DMOAP can effectively induce the homeotropic alignment of LC. In the presence of Cd2+, Cd2+ can induce DNA to bend and become a 2 nm spherical structure, which can greatly disrupt the orientational arrangement of LC, resulting in the correspond changes of the optical image of LC cell birefringent under the polarizing microscope. When the Cd2+ concentration is low to 0.1 nM, the optical signal of LC biosensor has an obvious change. But in the absence of Cd2+, there is no orientational response of LC and the optical image under the polarizing microscope is still a uniform dark background. Thus, this LC sensing method has a sensitive and clear distinction between positive and negative results and offers a highly sensitive detection of Cd2+ with a low detection limit down to 0.1 nM.展开更多

In the absence o...In this report, we present a method for the detection of Pb2+ based on the different adsorption capacity on the surface of gold nanoparticles (AuNPs) between ssDNA (single-stranded DNA) and G-quartet. In the absence of Pb2+, the DNA oligonucleotides probe, which is guanine-rich ssDNA, can be adsorbed on the surface of AuNPs protecting them from aggregation. After adding Pb2+, the DNA oligonucleotides probe can specifically form compact G-quartet, which can induce the aggregation of unmodified AuNPs, especially after adding NaCl aqueous solution. Consequently, the color turns from red to blue. Pb2+ can be detected by colorimetric response of AuNPs;its detection limit can reach 5 μM only observed by naked eyes. Most metal ions have no interferences, and the interference of Cu2+ can be effectively eliminated by adding cysteine. It provides a simple and effective colorimetric sensor for on-site and real time detection of Pb2+.展开更多

The rats we...This study was conducted to investigate the gene expression in fructose-fed rat skeletal muscle by cDNA chip which could provide support to elucidate the molecular mechanisms underlying insulin resistance. The rats were divided into two groups, one of which was normal control and the other was fed with fructose-rich diet. The mRNA was isolated and purified from the skeletal muscle of two groups. The mRNA from two kinds of tissue was reverse transcribed to cDNA with Cy3-dUTP and Cy5-dUTP separately to prepare hybridization probes. The mixed probes were hybridized to cDNA microarray. The microarray was scanned, analyzed and repeated for two times. Among the total 4 096 tested genes, 140 genes were differently expressed, 62 up-regulated,78 down-regulated, the expression of Ptprd and Gilz and multiple genes of oxidative metabolism is associated in insulin resistance. The differential expression of gene may be related to the pathogenesis of insulin resistance.展开更多
Purpose:To explore the application effect of the cultivation mode of Curriculum Ideology and Political Integration of Medical Humanities to enhance the core values of medicine in the clinical internship stage of medic...Purpose:To explore the application effect of the cultivation mode of Curriculum Ideology and Political Integration of Medical Humanities to enhance the core values of medicine in the clinical internship stage of medical students.Methods:Students in the clinical internship stage of the First Affiliated Hospital of Baotou Medical College were selected,and a total of 156 students in one random class in each grade were taken as the observation group and received the integration cultivation mode;148 students were taken as the control group and employed the traditional mode.The teaching effect of interns in the two groups was analyzed.Results:The teaching performance of students in both groups after clinical internship teaching was improved compared with that before admission;the teaching performance,teaching effect,teaching evaluation,and teaching satisfaction of the observation group were higher than that of the control group.Conclusion:The integration of the medical humanities training model with curriculum ideology and politics in the clinical internship of medical students is conducive to the improvement of teaching performance,teaching evaluation,teaching satisfaction of teachers and students,and the development and improvement of core values of medical students,which is of good value for teaching application.展开更多
Background and Aims:Hepatitis B virus(HBV)reactivation is commonly observed in individuals with chronic HBV infection undergoing antineoplastic drug therapy.Paclitaxel(PTX)treatment has been identified as a potential ...Background and Aims:Hepatitis B virus(HBV)reactivation is commonly observed in individuals with chronic HBV infection undergoing antineoplastic drug therapy.Paclitaxel(PTX)treatment has been identified as a potential trigger for HBV reactivation.This study aimed to uncover the mechanisms of PTX-induced HBV reactivation in vitro and in vivo,which may inform new strategies for HBV antiviral treatment.Methods:The impact of PTX on HBV replication was assessed through various methods including enzyme-linked immunosorbent assay,dual-luciferase reporter assay,quantitative real-time PCR,chromatin immunoprecipitation,and immunohistochemical staining.Transcriptome sequencing and 16S rRNA sequencing were employed to assess alterations in the transcriptome and microbial diversity in PTX-treated HBV transgenic mice.Results:PTX enhanced the levels of HBV 3.5-kb mRNA,HBV DNA,HBeAg,and HBsAg both in vitro and in vivo.PTX also promoted the activity of the HBV core promoter and transcription factor AP-1.Inhibition of AP-1 gene expression markedly suppressed PTX-induced HBV reactivation.Transcriptome sequencing revealed that PTX activated the immune-related signaling networks such as IL-17,NF-κB,and MAPK signaling pathways,with the pivotal common key molecule being AP-1.The 16S rRNA sequencing revealed that PTX induced dysbiosis of gut microbiota.Conclusions:PTX-induced HBV reactivation was likely a synergistic outcome of immune suppression and direct stimulation of HBV replication through the enhancement of HBV core promoter activity mediated by the transcription factor AP-1.These findings propose a novel molecular mechanism,underscoring the critical role of AP-1 in PTX-induced HBV reactivation.展开更多
Although neuroelectrochemical sensing technology offers unique benefits for neuroscience research,its application is limited by substantial interference in complex brain environments while ensuring biosafety requireme...Although neuroelectrochemical sensing technology offers unique benefits for neuroscience research,its application is limited by substantial interference in complex brain environments while ensuring biosafety requirements.In this study,we introduced poly(3-hexylthiophene)(P3HT)and nitrogen-doped multiwalled carbon nanotubes(N-MWCNTs)to construct a composite membrane-modified carbon fiber microelectrode(CFME/P3HT-N-MWCNTs)for ascorbic acid(AA)detection.The microelectrode presented good linearity,selectivity,stability,antifouling,and biocompatibility and exhibited great performance for application in neuroelectrochemical sensing.Subsequently,we applied CFME/P3HT-N-MWCNTs to monitor AA release from in vitro nerve cells,ex vivo brain slices,and in vivo living rat brains and determined that glutamate can induce cell edema and AA release.We also found that glutamate activated the N-methyl-d-aspartic acid receptor,which enhanced Na^(+) and Cl^(−) inflow to induce osmotic stress,resulting in cytotoxic edema and ultimately AA release.This study is the first to observe the process of glutamate-induced brain cytotoxic edema with AA release and to reveal the mechanism.Our work can benefit the application of P3HT in in vivo implant microelectrode construction to monitor neurochemicals,understand the molecular basis of nervous system diseases,and discover certain biomarkers of brain diseases.展开更多

文摘Background and Aims:Hepatitis B virus(HBV)reactivation is commonly observed in individuals with chronic HBV infection undergoing antineoplastic drug therapy.Paclitaxel(PTX)treatment has been identified as a potential trigger for HBV reactivation.This study aimed to uncover the mechanisms of PTX-induced HBV reactivation in vitro and in vivo,which may inform new strategies for HBV antiviral treatment.Methods:The impact of PTX on HBV replication was assessed through various methods including enzyme-linked immunosorbent assay,dual-luciferase reporter assay,quantitative real-time PCR,chromatin immunoprecipitation,and immunohistochemical staining.Transcriptome sequencing and 16S rRNA sequencing were employed to assess alterations in the transcriptome and microbial diversity in PTX-treated HBV transgenic mice.Results:PTX enhanced the levels of HBV 3.5-kb mRNA,HBV DNA,HBeAg,and HBsAg both in vitro and in vivo.PTX also promoted the activity of the HBV core promoter and transcription factor AP-1.Inhibition of AP-1 gene expression markedly suppressed PTX-induced HBV reactivation.Transcriptome sequencing revealed that PTX activated the immune-related signaling networks such as IL-17,NF-κB,and MAPK signaling pathways,with the pivotal common key molecule being AP-1.The 16S rRNA sequencing revealed that PTX induced dysbiosis of gut microbiota.Conclusions:PTX-induced HBV reactivation was likely a synergistic outcome of immune suppression and direct stimulation of HBV replication through the enhancement of HBV core promoter activity mediated by the transcription factor AP-1.These findings propose a novel molecular mechanism,underscoring the critical role of AP-1 in PTX-induced HBV reactivation.

文摘Although neuroelectrochemical sensing technology offers unique benefits for neuroscience research,its application is limited by substantial interference in complex brain environments while ensuring biosafety requirements.In this study,we introduced poly(3-hexylthiophene)(P3HT)and nitrogen-doped multiwalled carbon nanotubes(N-MWCNTs)to construct a composite membrane-modified carbon fiber microelectrode(CFME/P3HT-N-MWCNTs)for ascorbic acid(AA)detection.The microelectrode presented good linearity,selectivity,stability,antifouling,and biocompatibility and exhibited great performance for application in neuroelectrochemical sensing.Subsequently,we applied CFME/P3HT-N-MWCNTs to monitor AA release from in vitro nerve cells,ex vivo brain slices,and in vivo living rat brains and determined that glutamate can induce cell edema and AA release.We also found that glutamate activated the N-methyl-d-aspartic acid receptor,which enhanced Na^(+) and Cl^(−) inflow to induce osmotic stress,resulting in cytotoxic edema and ultimately AA release.This study is the first to observe the process of glutamate-induced brain cytotoxic edema with AA release and to reveal the mechanism.Our work can benefit the application of P3HT in in vivo implant microelectrode construction to monitor neurochemicals,understand the molecular basis of nervous system diseases,and discover certain biomarkers of brain diseases.
