真空低温慢煮对猪肉肠品质特性的影响

为探明真空低温慢煮在猪肉肠加工中的应用潜力,以传统蒸煮(100℃、30 min)为对照,研究不同加热温度(60、65、70℃)和时间(0、1、2、3、4 h)真空低温慢煮处理对猪肉肠蒸煮损失率、pH值、色泽、质构特性、剪切力、营养成分、菌落总数及感... 为探明真空低温慢煮在猪肉肠加工中的应用潜力,以传统蒸煮(100℃、30 min)为对照,研究不同加热温度(60、65、70℃)和时间(0、1、2、3、4 h)真空低温慢煮处理对猪肉肠蒸煮损失率、pH值、色泽、质构特性、剪切力、营养成分、菌落总数及感官特性的影响。结果表明,随着加热温度升高或加热时间的延长,猪肉肠蒸煮损失率、pH值、红度值和黄度值呈上升趋势,亮度值相对稳定,剪切力总体呈先上升再下降的趋势;蛋白质和脂肪等营养品质无明显损失,菌落总数降低且均符合熟肉制品国家标准要求。与传统蒸煮对比,真空低温慢煮可有效提高产品得率、减少水分流失、改善质构特性、提高嫩度,感官品质更优。主成分分析所提取的4个主成分累计方差贡献率达73.582%,综合评价模型结果显示,70℃真空低温慢煮3 h猪肉肠的综合品质更优。 展开更多

An accurate assessment of host and pathogen gene expression during infection is critical for understanding the molecular aspects of host-pathogen interactions.Often,pathogen-derived transcripts are difficult to ascertain at early infection stages owing to the unfavourable transcript representation compared to the host genes.In this study,we compare two sequencing techniques,RNAseq and enrichment sequencing(RenSeq and PenSeq)of cDNA,to investigate gene expression patterns in the doubled monoploid potato(DM)infected with the late blight pathogen Phytophthora infestans.Our results reveal distinct advantages of cDNA RenSeq and PenSeq over traditional RNAseq in terms of target gene representation and transcriptional quantification at early infection stages.Throughout the infection time course,cDNA enrichment sequencing enables transcriptomic analyses for more targeted host and pathogen genes.For highly expressed genes that were sampled in parallel by both cDNA enrichment and RNAseq,a high level of concordance in expression profiles is observed,indicative of at least semi-quantitative gene expression representation following enrichment. 展开更多

The microbial community and flavour substances of off-flavoured rice sour soup during storage

Fermented sour soup is a unique traditional sour food of the Miao and Dong ethnic groups in China.However,due to the commonly used traditional fermentation,the unstable fluctuation of microbial community often leads to some unpleasant inherent off-flavours in rice sour soup.In this study,the microbial composition and volatile flavour components of off-flavour and normal rice sour soup were examined by high-throughput sequencing and headspace solid-phase microextraction coupled with gas chromatography mass spectrometry.Then,based on Pearson correlation coefficients,the correlations between fungi and bacteria and those between core microorganisms and key volatile flavour components were investigated.The dominant fungal genera included Candida,Dekkera,Pichia,Rhizopus,and Issatchenkia,whereas the dominant bacterial genera included Lactobacillus,Pectinatus,Bifidobacterium,Pseudomonas,and Acetobacter.Compared to normal rice sour soup,off-flavoured rice sour soup contained the significantly increased relative abundance of Candida and the significantly decreased relative abundance of Dkkera.In addition,80 volatile flavour compounds detected in off-flavoured rice sour soup,mainly including esters,acids,and alcohols,were different from those detected in normal rice sour soup.Especially,some substances were only detected in off-flavoured rice sour soup,such as 1-hexanol(68-207μg/kg),butyric acid(20-92μg/kg)and heptanoic acid(12-45μg/kg),which might be the main source of off-flavours.These results provide new ideas and strategies of removing or abating odour from fermented rice sour soup. 展开更多

Irritable bowel syndrome-diarrhea(IBS-D)is a prevalent functional bowel disease severely impairs the quality of life of patients.Visceral hypersensitivity(VH)is the main symptom of IBS-D.Probiotics have been found to effectively alleviate VH.In this study,neonatal maternal separation(NMS)was used to induce VH in 72 rats and investigated the effects of daily gavage of 3×10^(9) CFU of Lactobacillus and Bifidobacterium on alleviating the symptoms of IBS-D and behavioral interventions,as well as exploring potential mechanisms.The results demonstrated that L.acidophilus and B.breve could alleviate the symptoms of IBS-D such as VH and increased frequency of bowel movements,as well as improving non-spatial memory in NMS rats.These effects were achieved by modulating the composition of gut microbiota and metabolites(including short-chain fatty acids,deoxycholic acid and adenosine),and by regulating the expression of genes and proteins associated with the NGF/TrkA signaling pathway,tryptophan metabolism,and inflammatory factors in the hippocampus.This finding suggests that L.acidophilus and B.breve have promising potential for the alleviation of IBS. 展开更多

This study endeavors to investigate the effects of Bifidobacterium breve CCFM1078 on bone formation and resorption balance in growing BALB/c mice.Newborn BALB/c mice were assigned to the control group(administration saline)and the CCFM1078 group(administration B.breve CCFM1078,3×10^(9) CFU/day)in 3-,4-,and 5-week tests.All the groups have male and female distinctions.Our findings demonstrate that B.breve CCFM1078 exerts on the dynamic equilibrium between bone formation and resorption during the critical period of growth in mice by modulating the composition of gut microbiota and metabolites(hexadecanamide,linoleoyl ethanolamide,and palmitoyl ethanolamide),the genes and proteins expression related to the growth hormone(GH)/insulin-like growth factors-1(IGF-1)axis and Gs/PKA/CREB signaling pathways,as well as downstream osteogenic and osteoclastic differentiation factors.The effects of B.breve CCFM1078 were different with age and gender dependent.This finding suggests B.breve CCFM1078 may have potential applications in regulating bone metabolism in the growth period population. 展开更多

Bifidobacterium longum subsp.infantis is a commensal bacterium that predominates in the infant gut,playing a critical role in both preventing foreign infections and facilitating immune development.This study aimed to explore the effects of B.longum subsp.infantis supplementation on interferon-beta(IFN-β)secretion and intestinal barrier improvement in growing mice.Female and male mice were orally administered either saline or B.longum subsp.infantis CCFM1269 or I5TI(1×10^(9) CFU/mice per day,n=8)from 1-week-age until 3-,4-,and 5-week-age.RNA sequencing analysis revealed that CCFM1269 exhibited potential antiviral capacity through increasing 2'-5'oligoadenylate synthetase(OAS).Additionally,CCFM1269 supplementation significantly increased colonic IFN-β levels which combined with OAS in 3-week-old female and male mice by activating the TLR4-TRIF-dependent signaling pathway.However,this effect was not observed in 4-and 5-week-old mice.Furthermore,both CCFM1269 were found to modulate the gut microbiota composition and enhance the intestinal barrier function in 3-,4-,and 5-week-old mice.In summary,the results of this study suggested that B.longum subsp.infantis CCFM1269 promoting intestinal barrier and releasing IFN-β in growing mice was in a strain-specific and time-dependent manner. 展开更多

为了进一步拓展海洋鱼类蛋白在食品中的应用,以蓝圆鲹分离蛋白为原料,利用酶解改性得到溶解性良好的蓝圆鲹分离蛋白酶解物(BPIH),并研究其对大米淀粉(RS)短期老化的抑制作用。通过响应面法优化酶解改性工艺制备BPIH,并分别按RS的3%、6%... 为了进一步拓展海洋鱼类蛋白在食品中的应用,以蓝圆鲹分离蛋白为原料,利用酶解改性得到溶解性良好的蓝圆鲹分离蛋白酶解物(BPIH),并研究其对大米淀粉(RS)短期老化的抑制作用。通过响应面法优化酶解改性工艺制备BPIH,并分别按RS的3%、6%、9%(质量百分比)进行添加。通过测定RS的凝沉性、动态黏弹性、热学特性以及微观结构分析评价BPIH的抗淀粉老化活性。结果显示,响应面法确定的最佳条件:酶底比5000;1(U/g)、酶解时间3 h、料液比1.00;3.81、酶解温度46.36℃、酶解pH 6.30,氮溶指数(NSI)实际值为85.41%±0.82%,与预测值86.37%接近。该酶解条件下BPIH水解度达到21.62%。BPIH中小于1000 u的肽占79.94%,主要为小分子低聚寡肽。加入BPIH可以减弱RS的凝沉现象,降低RS在4℃保存过程中的储能模量(G'),显著降低老化后RS的峰值温度(T_(p))和焓值(ΔHr);加入BPIH的大米淀粉,老化后微观结构有较大孔洞,提升了淀粉糊化后保留内部水分的能力。研究表明,BPIH能够抑制RS在4℃保存期间凝胶网络和微晶结构的形成,同时可能限制了淀粉分子间的聚集,抑制或延缓RS的短期老化。本研究为蓝圆鲹分离蛋白酶解物在食品蛋白配料中的应用提供理论参考。 展开更多

为研究不同干燥方式对低盐腌制鲍鱼复水品质的影响,本研究采用真空冷冻干燥、冷风干燥与热风干燥对其进行干燥处理,研究其复水过程的质量体积和复水率,并通过低场核磁共振(low field nuclear magnetic resonance,LF-NMR)和磁共振成像(ma... 为研究不同干燥方式对低盐腌制鲍鱼复水品质的影响,本研究采用真空冷冻干燥、冷风干燥与热风干燥对其进行干燥处理,研究其复水过程的质量体积和复水率,并通过低场核磁共振(low field nuclear magnetic resonance,LF-NMR)和磁共振成像(magnetic resonance imaging,MRI)研究复水过程的水分分布以及复水后的色差、质构、组织形态、游离氨基酸等变化情况。结果表明,在72 h时真空冷冻干鲍的复水率为278.73%,显著高于冷风干鲍和热风干鲍的复水率(P<0.05)。由LF-NMR和MRI结果分析得出,不易流动水和自由水含量是三种干鲍水分含量增加的主要因素。真空冷冻干鲍、冷风干燥和热风干鲍分别在72、48和24 h时复水完成。不同的干燥方式影响复水后的质构,结果显示,三种干燥方式的鲍鱼样品硬度与咀嚼性均有显著差异(P<0.05),真空冷冻干燥鲍鱼复水后硬度和咀嚼性最低,分别为954.01和708.59 g;热风干燥鲍鱼复水后硬度与咀嚼性最高,分别为1230.14和920.02 g。组织学染色与扫描电镜结果显示,真空冷冻干鲍的肌肉组织为较疏松的多孔结构;冷风干鲍和热风干鲍的组织为较细长和致密的结构。综上,真空冷冻干燥鲍鱼的复水率最高,硬度和咀嚼性最低,口感最好,但滋味不及热风干燥鲍鱼。本研究结果可为了解不同干燥方法对低盐腌制鲍鱼产品品质的影响提供理论参考。 展开更多

Globally,sub-optimal use of nitrogen (N) fertiliser and elevated N irrigation groundwater have led to high leached nitrate (NO_(3)^(–)) losses from protected vegetable field systems.Optimising fertiliser and irrigation management in different soil types is crucial to reduce future N loads from such systems.The present 4-year study examined leached N loads from lysimeter monitoring arrays set up across 18 protected vegetable system sites encompassing the dominant soil types of northern China.The treatments applied at each field site were:1) a high N and high irrigation input treatment (HNHI);2) a low N but high irrigation input treatment (LNHI) and 3) a low N with low irrigation input treatment (LNLI).Results showed that the mean annual leached total nitrogen loads from the HNHI,LNHI and LNLI treatments were 325,294 and 257 kg N ha^(–1) in the fluvo-aquic soil,114,100 and 78 kg N ha^(–1) in the cinnamon soil and 79,68 and 57 kg N ha^(–1) in the black soil,respectively.The N dissolved in irrigation water in the fluvo-aquic soil areas was 8.26-fold higher than in the cinnamon areas.A structural equation model showed that N fertiliser inputs and leaching water amounts explained 14.7 and 81.8%of the variation of leached N loads,respectively.Correspondingly,reducing irrigation water by 21.5%decreased leached N loads by 20.9%,while reducing manure N and chemical N inputs by 22 and 25%decreased leached N loads by only 9.5%. This study highlights that protected vegetable fields dominated by fluvo-aquic soil need management to curtail leached N losses in northern China. 展开更多

With methane emissions from ruminant agriculture contributing 17%of total methane emissions worldwide,there is increasing urgency to develop strategies to reduce greenhouse gas emissions in this sector.One of the proposed strategies is ruminant feed intervention studies focused on the inclusion of anti-methanogenic compounds which are those capable of interacting with the rumen microbiome,reducing the capacity of ruminal microorganisms to produce methane.Recently,seaweeds have been investigated for their ability to reduce methane in ruminants in vitro and in vivo,with the greatest methane abatement reported when using the red seaweed Asparagopsis taxiformis(attributed to the bromoform content of this species).From the literature analysis in this study,levels of up to 99%reduction in ruminant methane emissions have been reported from inclusion of this seaweed in animal feed,although further in vivo and microbiome studies are required to confirm these results as other reports showed no effect on methane emission resulting from the inclusion of seaweed to basal feed.This review explores the current state of research aiming to integrate seaweeds as anti-methanogenic feed additives,as well as examining the specific bioactive compounds within seaweeds that are likely to be related to these effects.The effects of the inclusion of seaweeds on the ruminal microbiome are also reviewed,as well as the future challenges when considering the largescale inclusion of seaweeds into ruminant diets as anti-methanogenic agents. 展开更多

Relating estimates of wood properties of birch to stem form, age and species

Birch has long suffered from a lack of active forest management,leading many researchers to use mate-rial without a detailed management history.Data collected from three birch(Betula pendula Roth,B.pubescens Ehrh.)sites in southern Sweden were analyzed using regression analysis to detect any trends or differences in wood proper-ties that could be explained by stand history,tree age and stem form.All sites were genetics trials established in the same way.Estimates of acoustic velocity(AV)from non-destructive testing(NDT)and predicted AV had a higher correlation if data was pooled across sites and other stem form factors were considered.A subsample of stems had radial profiles of X-ray wood density and ring width by year created,and wood density was related to ring number from the pith and ring width.It seemed likely that wood density was negatively related to ring width for both birch species.Linear models had slight improvements if site and species were included,but only the youngest site with trees at age 15 had both birch species.This paper indicated that NDT values need to be considered separately,and any predictive models will likely be improved if they are specific to the site and birch species measured. 展开更多

Lactobacillus are considered promising therapeutic methods for nonalcoholic fatty liver disease(NAFLD).The effects of two strains of Ltmosilactobacillus mucosae on NAFLD were investigated in this study.Fourweek-old male C57BL/6J mice were divided into 4 groups(n=8 per group,Control,Model,FZJTZ26M3,FGSYC17L3).L.mucosae FZJTZ26M3 reduced the mice 's body weight,liver weight,and adipose tissue weight after 12 weeks of therapy.According to serum analysis,total cholesterol,triacylglycerol,and low-density lipoprotein cholesterol significantly decreased after L.mucosae FZJTZ26M3 intervention.Liver pathology showed that L.mucosae FZJTZ26M3 was effective to ameliorate lipid deposition in NAFLD mice.Additionally,the expression of the gene related to lipid metabolism in the liver and adipose tissue was analyzed,and the results indicated that L.mucosae FZJTZ26M3 could alleviate NAFLD by regulating lipid metabolism.Furthermore,the results of 16S rRNA gene sequencing revealed a drop in the relative abundance of Ruminococcaceae,which is linked to inflammation,but the relative abundance of a potential probiotic Akkermansia significantly increased after L.mucosae FZJTZ26M3 intervention.Generally,L.mucosae FZJTZ26M3 could be a candidate to prevent NAFLD. 展开更多

This study aimed to evaluate the effects of Bifi dobacterium breve CCFM683 on psoriasis and to investigate the underlying mechanisms.B.breve CCFM683 significantly ameliorated psoriasis in mice as well as elevated the deoxycholic acid(DCA)and lithocholic acid(LCA)in the colon compared with those of the imiquimod(IMQ)-treated mice.Meanwhile,B.breve CCFM683 increased the relative abundance of DCA-producing Lachnoclostridium and diminished the harmful Desulfovibrio and Prevotellaceae UCG001.Additionally,the farnesoid X receptor(FXR)in the skin was activated and the expression of the Toll-like receptor 4(TLR4)/nuclear factor kappa-B(NF-κB)pathway was inhibited,and the downstream interleukin(IL)-17 and tumor necrosis factor(TNF)-αwere downregulated whereas IL-10 was up-regulated.Moreover,the subsequent hyperproliferation of keratinocytes and the dysfunction of the epidermal barrier were improved.In conclusion,CCFM683 administration ameliorated IMQ-induced psoriasis via modulating gut microbiota,promoting the DCA production,regulating the FXR-TLR4/NF-κB pathway,diminishing proinflammatory cytokines,and regulating keratinocytes and epidermal barrier.These findings may be conducive to elucidating the mechanism for probiotics to ameliorate psoriasis and to promote its clinical trials in skin disease. 展开更多

为探究魔芋胶(konjac glucomannan,KGM)对南美白对虾肌原纤维蛋白(shrimp myofibrillar protein,SMP)凝胶性能的影响,将KGM与SMP按不同比例(1∶50、1∶20、1∶10)进行复配制备复合凝胶体系SK50、SK20、SK10,通过测定表面疏水性、内源性... 为探究魔芋胶(konjac glucomannan,KGM)对南美白对虾肌原纤维蛋白(shrimp myofibrillar protein,SMP)凝胶性能的影响,将KGM与SMP按不同比例(1∶50、1∶20、1∶10)进行复配制备复合凝胶体系SK50、SK20、SK10,通过测定表面疏水性、内源性荧光、浊度及粒径、流变学、红外光谱、蛋白变化及凝胶微观结构,研究复合体系的凝胶特性变化。结果表明,SMP及SMP-KGM复合体系的表面疏水性、浊度及粒径随着KGM添加量的增大而增大,流变学分析表明SMP及SMP-KGM复合体系均出现剪切稀化现象,且SK20的G’值最高,其凝胶的结构稳定性最好。红外光谱分析表明SMP及SMP-KGM复合体系组的光谱特征带相似,表明无明显的基团生成。十二烷基硫酸钠-聚丙烯酰胺凝胶电泳分析表明KGM的加入并没有引起蛋白条带的明显变化,相较于未加热组,加热后样品中肌球蛋白重链、副肌球蛋白以及肌动蛋白的条带明显减弱,说明加热能促使三者发生热聚合反应。扫描电镜与分形维数分析表明,随着KGM添加量的增加,形成了致密有序的凝胶。结果表明,一定量的KGM添加能有效提升虾糜的凝胶特性,提高虾糜类产品品质。 展开更多

以荞麦(Fagopyrum esculentum Moench.)为原料,通过碱溶酸沉法提取荞麦分离蛋白(BPI),分别采用真空冷冻干燥与喷雾干燥制备蛋白粉,研究谷氨酰胺转氨酶(TGase)及不同干燥方式对荞麦分离蛋白理化性质和凝胶特性的影响。结果表明,冷冻干燥... 以荞麦(Fagopyrum esculentum Moench.)为原料,通过碱溶酸沉法提取荞麦分离蛋白(BPI),分别采用真空冷冻干燥与喷雾干燥制备蛋白粉,研究谷氨酰胺转氨酶(TGase)及不同干燥方式对荞麦分离蛋白理化性质和凝胶特性的影响。结果表明,冷冻干燥荞麦分离蛋白(FBPI)的蛋白组成与BPI类似,而喷雾干燥荞麦分离蛋白(SBPI)的13S球蛋白发生降解。FBPI和SBPI的α-螺旋含量分别为22.10%和20.00%,β-转角含量为19.40%和19.80%,表明喷雾干燥会使蛋白结构更加无序化。SBPI的表面疏水性指数显著(P<0.05)高于FBPI与BPI。与BPI相比,SBPI具有更高的表面疏水性和更均匀的粒径,但溶解度和持水能力较低,而FBPI具有与BPI相似的物理化学性质。加入TGase后,FBPI和SBPI的平均粒径分别由213.01与192.55 nm增加至2289.01与1439.67 nm,表明发生了交联反应。BPI、FBPI和SBPI的电位分别为−15.93、−29.43、−29.35 mV,表明干燥处理能进一步提高蛋白的溶液稳定性。相比于SBPI,BPI和FBPI在升温过程中G'持续下降,表明热处理会破坏BPI和FBPI的蛋白凝胶结构。经加热处理再进行冷却,FBPI的G'显著(P<0.05)高于SBPI与BPI,具有更好的凝胶性。TGase能显著(P<0.05)提高FBPI的储能模量与热稳定性,但对SBPI作用不明显。化学作用力结果显示,疏水相互作用和二硫键是凝胶主要作用力;TGase能有效促进凝胶结构中疏水相互作用和二硫键形成,但会导致蛋白分子间氢键断裂。由此可见,通过冷冻干燥制备的荞麦蛋白能在TGase的作用下形成凝胶性与稳定性较好的凝胶。本研究的结果为荞麦分离蛋白凝胶特性研究及其凝胶类食品的开发与应用提供理论参考。 展开更多

为探究干燥方式对蛋清蛋白(egg white protein,EWP)功能特性的影响及其内在机理,分别通过喷雾干燥与真空冷冻干燥制备蛋清蛋白粉,并对其蛋白结构、理化性质与功能特性进行研究。结果表明,与蛋清液(EWPC)相比,喷雾干燥使蛋清蛋白(EWP-P)... 为探究干燥方式对蛋清蛋白(egg white protein,EWP)功能特性的影响及其内在机理,分别通过喷雾干燥与真空冷冻干燥制备蛋清蛋白粉,并对其蛋白结构、理化性质与功能特性进行研究。结果表明,与蛋清液(EWPC)相比,喷雾干燥使蛋清蛋白(EWP-P)的内源性荧光强度降低,表面疏水性和表面游离巯基含量增大。傅里叶变换红外光谱分析显示,EWP-P的α-螺旋、β-折叠和β-转角分别为16.30%、25.72%和40.23%,冷冻干燥蛋清蛋白(EWP-D)分别为20.43%、24.32%和35.69%。不同pH下,EWP-D的溶解度均高于EWP-P,表面张力小于EWP-P。此外,EWP-P的接触角为99.62°,高于EWP-D(接触角为65.97°),表明喷雾干燥能显著提高蛋白的疏水性(P<0.05)。EWP-D在不同pH下的乳化性、乳化稳定性以及起泡性均大于EWP-P,但起泡稳定性更小,这与EWP-D较高的溶解性与较低的表明疏水性有关。荧光倒置显微镜及激光共聚焦扫描显微镜分析表明EWP-D乳液的微粒更小,分布更均匀,其稳定性高于EWP-P。综上,喷雾干燥蛋清蛋白的β-折叠结构较多,表面游离巯基含量和表面疏水性较高,具有较好的凝胶性;冷冻干燥蛋清蛋白的表面疏水性较小,且表面张力小、溶解度大,具有更好的乳化能力与起泡性。 展开更多
