SEM, EDAX, XRD, AFM and Scratch Testers are adopted to conduct analysis and research on the microstructure, composition, phase and bonding strength of the coating. Results indicate that the microstructure of coating is metallurgically bonded with stainless steel base, eliminating porosities and cracks. The coating has a considerable quantity of nanometer particles visible with a granularity ^lOOnm under a nanoscope atomic microscope. The bonding strength of the laser cladded coating is remarkably improved respectively compared with conventional hot-sprayed coating and spray welding. The nanometer effect of nanometer WC/Co introduced into the coating plays an important role in the laser cladding processes.展开更多

METHODS: cDNA fragments encoding M2-reactive epitopes of pyruvate dehydrogenase complex Ez (PDCE2), branched chain 2-oxo-acid dehydrogenase complex E2 (BCOADC-E2) and 2-oxo-glutarate dehydrogenase complex E2 (OGDC-E2) were amplified with PCR using total RNA extracted from human peripheral mononuclear blood cells. The fragments were cloned into the plasmid vector pQE-30 and then transferred into E. coliM15 (pREP4) for expression, which was induced by isopropylthio-β-Dgalactoside. The expressed recombinant BPO protein was demonstrated by SDS-PAGE, Western-blotting and Immunoabsorption test, its antigenic reactivity and specificity were identified with seven M2-positive sera confirmed at Euroimmun Research Center (Germany).Using the purified BPO, M2 antibodies in sera from patients with PBC and other liver related diseases were detected with ELISA.
RESULTS: The expressed BPO was observed with both antigenic reactivity and specificity of M2 autoantigens. The determination of M2 antibodies by BPO with ELISA was more sensitive than using the Euroimmun's kit with the coefficients of variation less than 10 % in both interassay and intraassay.With the newly established method, M2 antibodies were found in 100 % (20/20) of patients with PBC. Six cases of liver disease with unknown etiology and 1 patient with drug induced liver injury had detectable levels of serum M2antibodies. There were also 2 patients with autoimmune cholangitis and 1 with autoimmune hepatitis showing M2-antibody positive.
CONCLUSION: Compared with the routine immunofluorescenoe assay and commercially available assay kit using porcine heart mitochondrial protein as the antigen, the detection system established in the present study shows higher sensitivity and specificity and may be used as a powerful tool for the diagnosis of PBC.展开更多

Effects of Fe ion implantation on microstructure and friction coefficient of the DLC were studied. With increasing Fe ion fluence, friction coefficient of the DLC film increased as compared with that of DLC without implantation, and then decreased. The Raman spectra characteristics also show a dependence on the Fe ion fluence. With increasing the ion fluence, the sp2 bonding increased in the DLC film, resulting in the decrease of friction coefficient of the film af- ter implantation. The present study provides an extensionby using a hysteretic spring friction model and taking account of the stick-slip motion of theplate. Results for a range of problem parameters have been obtained. The results show that thenonlinear frequency response behavior of the system can be quite different from the rigid-interfacefriction model. The effects of the stiffness at friction interfaces and the stick-slip motion on thenonlinear vibration of the plate are significant and hence cannot be neglected.展开更多

The issues associated with tool management include tool requirement planning, tool real-time scheduling, tool crib management, tool inventory control, tool fault diagnosis, tool tracking and tool monitoring. In order to make tools flow into/out of FMS efficiently, this work is aimed to design a knowledge-based decision support system (KBDSS) for tool management in FMS. Firstly an overview of tool management functions is described. Then the structure of KBDSS for tool management and the essential agents in the design of KBDSS are presented.
